摘 要:SVP转录因子在调控植物生长发育以及应答非生物胁迫过程中发挥着重要作用。本研究通过同源克隆和RACE技术在艾纳香(Blumea balsamifera)中获得了一个SVP-like基因,并命名为BbSVP。生物信息学分析表明,BbSVP基因的编码区长度为681 bp,编码226个氨基酸,属于MADS-box蛋白家族,蛋白内部无明显信号肽和跨膜结构域。同源性比对显示,BbSVP与同为菊科植物向日葵HaSVP X1的亲缘关系最近。进一步研究发现,BbSVP集中分布于细胞核。BbSVP基因在艾纳香中具有组织表达特异性,在叶片中表达量最高,在花中最低。对艾纳香叶片进行非生物胁迫处理后,RT-qPCR分析显示,BbSVP在转录水平上能够应答PEG、NaCl和4℃低温胁迫。研究表明,BbSVP基因在序列和功能上具有一定的保守性,其可能参与调控艾纳香响应非生物胁迫的过程。SVP(Short vegetative pase) transcription factor plays an important role in regulating plant growth and development and responding to abiotic stress. In this study, an SVP-like gene was obtained from Blumea balsamifera by homologous cloning and RACE techniques, which was named as BbSVP. Bioinformatics analysis showed that the coding region of the BbSVP gene was 681 bp long and encoded 226 amino acids, belonging to the MADS-box protein family, and there were no obvious signal peptides and transmembrane domains in the protein. Homology comparison showed that BbSVP was closely related to HaSVP X1 of sunflower, which was also in the asteraceae family. Further studies showed that BbSVP was concentrated in the nucleus. BbSVP gene had tissue specific expression, and the expression level of BbSVP gene was highest in leaves and lowest in flowers. After abiotic stress treatment on Blumea balsamifera,RT-qPCR analysis showed that BbSVP could respond to PEG, NaCl and 4 ℃ low temperature stress at the transcriptional level. The results show that BbSVP gene was conserved in sequence and function, and it could be involved in regulating the response to abiotic stress.
