摘 要:目的:观察助阳舒心方对抑郁大鼠的行为学影响,并通过海马组织脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)变化探究其作用机制。方法:50只SD大鼠随机分为正常组、模型组和助阳舒心方低、中、高剂量组,每组10只。采用单只单笼饲养加不可预见性慢性刺激的方法复制抑郁模型,给药21 d,观察大鼠体质量变化,进行旷场实验和糖水偏好实验,实时荧光定量PCR法和免疫组化法检测大鼠脑部海马组织BDNF水平。结果:与正常对照组对比,模型组大鼠体质量增长缓慢,在中心格停留增多且持续时间延长,垂直运动次数减少,糖水偏好率下降,免疫组化法和实时荧光定量PCR法均显示海马组织BDNF水平存在一定程度的下降;与模型组比较,各给药组体质量增速提高,对周围环境探索欲望增强,糖水偏好率上升,脑BNDF水平均呈上升趋势。结论:助阳舒心方改善不可预见性慢性抑郁模型大鼠的症状可能与提高脑内BDNF水平有关。Objective:To observe the effect of Zhuyang Shuxin Prescription on the behavior of depressed rats,and to explore its anti-depression mechanism by detecting the BDNF level in hippocampus.Methods:50 SD rats were randomly divided into normal group,model group,Zhuyang Shuxin Prescription low,moddle,high dose(2、4、8 g/kg)groups with each10 rats.CUMS and solitary culture were used to induce depression model for 21 days.The change of rats’body weight were recorded,open field test and sugar favouritism test were applied to observe rats’behavioral changes.The levels of BDNF in hippocampus were measured by real-time PCR and IHC.Results:Compared with the normal control group,the growth rate of body weight in the model group was decreased,rats stayed in the central grid for longer time,the number of rats’vertical movements was decreased,and the percentage of sugar water preference was decreased.Both IHC and real-time PCR showed that the level of BDNF in the hippocampus was decreased;Compared with the model group,the growth rate of body weight of the three dose Zhuyang Shuxin Prescription groups was increased,the desire to explore the surrounding environment was enhanced,the percentage of preference for sugar water was increased,and the BDNF level was increased.Conclusion:The improvement of symptoms of chronic unpredictable depressed model rats by Zhuyang Shuxin Prescription may be related to the increase of BDNF level in the hippocampus.
