摘 要:平邑甜茶(Malus hupehensis)作为砧木,被广泛应用于苹果生产。为进一步提高平邑甜茶的离体快繁效率,以平邑甜茶春季幼嫩新梢带芽茎段为外植体,探究不同外植体消毒方式、不同植物生长调节剂配比对组培苗增殖和生根培养的影响,建立平邑甜茶离体快繁体系。结果表明,平邑甜茶新梢带芽茎段用75%酒精处理30 s+2%次氯酸钠溶液处理12 min的消毒效果较好,成活率达81.11%。最佳增殖培养基为MS+1.0 mg/L 6-BA+0.1 mg/L IBA,增殖系数为7.09;最适生根培养基为1/2MS+0.15 mg/L NAA,该处理下的生根率为75.55%,平均根系数量和长度分别为3.23条、3.57 cm;生根后的组培苗在蛭石∶珍珠岩∶营养土质量比为1∶1∶1基质中的移栽成活率为90%。该离体快繁体系,为周年培育平邑甜茶砧木苗提供了技术支持。As an excellent apple rootstock,Malus hupehensis is widely used in apple production.In order to further improve the efficiency of in vitro rapid propagation of M.hupehensis,the spring stem segments with buds of M.hupehensis were used as explants to explore the effects of different explants disinfection methods as well as different hormone ratios on the proliferation and rooting of tissue cultured seedlings.The results showed that the explants immersed in 75%alcohol for 30 seconds,and then in 2%sodium hypochlorite for 12 min,had the best disinfection effect,with a survival rate of 81.11%.The optimal proliferation medium was MS+1.0 mg/L 6-BA+0.1 mg/L IBA,with a proliferation coefficient of 7.09.In addition,the optimal rooting medium was 1/2 MS+0.15 mg/L NAA with rooting rate of 75.55%,and the average number and length of roots were 3.23 and 3.57 cm,respectively.The survival rate of the transplanted tissue culture seedlings was 90%in substrate of vermiculite∶perlite∶nutrient soil=1∶1∶1.The in vitro rapid propagation system provided technical support for the year round cultivation of M.hupehensis rootstock seedlings.
