摘 要:为研究不同生长年限灵芝对杉木林下土壤微生物群落结构及其多样性的变化,并初步探索土壤因子与真菌、细菌群落的相关性,通过Illumina MiSeq高通量测序技术对杉木林下栽培1年(S1)、2年(S2)、3年(S3)、4年(S4)的灵芝栽培土壤进行了扩增子测序,分析了杉木林下不同生长年限灵芝栽培土壤中微生物群落的变化特征。结果表明,在门水平上,细菌主要优势菌群为变形菌门、酸杆菌门、放线菌门、绿弯菌门,其中变形菌门和酸杆菌门的优势表现较为显著;真菌优势菌群主要为子囊菌门、担子菌门、被孢霉门、罗兹菌门和一些未定种类,其中4组样品中子囊菌门占主导地位,相对丰度占比最高。在属水平上,细菌黄色杆菌属、norank_f__norank_o__Elsterales属相对丰度在4组样品中占比均较高,且4年相对丰度占比无显著差异;在真菌属水平上发现,每个年份的主要优势菌群均不同,S1组的主要优势菌群为木霉属,S2组的主要优势菌群为纹枯状革菌属,S3组的主要优势菌群为unclassified_k__Fungi属,S4组的主要优势菌群为unclassified_p_Basidiomycota属。土壤全碳、全磷、漆酶是影响杉木林下不同生长年限灵芝栽培土壤微生物群落结构和土壤微生物多样性的关键因子。In order to study the changes of Ganoderma lucidum with different growth years on the structure and diversity of soil microbial communities under fir forests,the correlation between soil factors and fungal and bacterial communities was preliminarily explored.Illumina MiSeq high-throughput sequencing technology was used to sequence the amplicon sequences of Ganoderma lucidum cultivation soil cultivated under fir forest for 1 year(S1),2 years(S2),3 years(S3)and 4 years(S4),and the characteristics of microbial community changes in Ganoderma lucidum cultivation soil under fir forest were analyzed.The results showed that at the level of the phylum,the main dominant bacterial floras are Proteobaceria,Acidobacteriota,Actinobacterota,and Chloroflexi,among which the advantages of Proteobaceria and Acidobacteria are more significant.The dominant fungal floras were Ascomycota,Basidiomycota,Mortierellomycota,Rozellomycota and unclassified fungi,among which the Ascomycota dominated the four groups of samples,and the relative abundance accounted for the highest proportion.At the genus level,the relative abundance of bacterial norank_f_Xanthobacteraceae and norank_f__norank_o__Elsterales genus was relatively high in the four groups of samples,and there was no significant difference in the proportion of relative abundance for four years.At the level of the genus of fungi,the dominant flora differs from year to year,and the S1 dominant flora is Trichoderma.The main dominant flora of S2 is Hypochnicium.The main dominant flora of S3 is unclassified_k__Fungi,and the dominant flora of S4 is the genus unclassified_p_Basidiomycota.Soil total carbon,total phosphorus and laccase were the key factors affecting the structure of soil microbial community and soil microbial diversity in Ganoderma lucidum cultivation under fir forests at different growth years.
