基于Runx2研究补肾类中药防治骨质疏松机制进展

摘　　要：研究发现Runx2在骨质疏松症(osteoporosis,OP)的发生发展过程中起到重要作用。Runx2通过结合特定的DNA序列来调节许多基因的转录,与骨代谢的关系密切,可通过多个信号通路、细胞因子、激素等对成骨细胞、软骨细胞、破骨细胞等具有调控作用。近年来,中医药在防治骨质疏松方面的研究越来越多,许多学者发现在骨质疏松症的治疗中补肾类中药与Runx2因子存在明显的相关性,如单药中的淫羊藿、骨碎补、杜仲、鹿茸、补骨脂,复方中的六味地黄丸、左归丸、补肾活血汤、二仙汤、仙灵骨葆胶囊,可通过作用于不同的信号通路,如Wnt/β-catenin、BMPs、MAPK、PI3K/AKT、Hedgehog等信号通路调控Runx2的表达以防治骨质疏松。故该文通过补肾类中药对Runx2因子调控相关信号通路防治骨质疏松症进行综述,以期为中药防治骨质疏松症的相关研究提供参考。It has been found that Runx2 plays an important role in the development of osteoporosis(OP).Runx2 regulates the transcription of many genes by binding to specific DNA sequences,which is closely related to bone metabolism,and can regulate osteoblasts,chondrocytes,osteoclasts and osteoclasts through multiple signaling pathways,cytokines,hormones,etc.In recent years,there have been more and more studies on the prevention and treatment of osteoporosis in Chinese medicine.In recent years,more and more researches on Chinese medicine in the prevention and treatment of osteoporosis have been conducted,and many scholars have found that there is an obvious correlation between kidney tonifying Chinese medicines and Runx2 factor in the therapeutic principle of osteoporosis,such as Yinyanghuo(Epimedii Wushanensis Folium),Gusuibu(Drynariae Rhizoma),Duzhong(Eucommiae Cortex),Lurong(Cervi Cornu Pantotrichum),Buguzhi(Psoraleae Fructus)in single medicine,and Liuwei Dihuang Pills(六味地黄丸),Zuogui Pills(左归丸),Bushen Huoxue Decoction(补肾活血汤),Erxian Decoction(二仙汤),and Xianling Gubao Capsules(仙灵骨葆胶囊)in compound medicine.The capsules can regulate the expression of Runx2 by acting on different signaling pathways,such as Wnt/β-catenin,BMP,MAPK,PI3K/AKT,Hedgehog,and so on.Therefore,in this paper,we review the regulation of Runx2 by kidney tonifying traditional Chinese medicine to prevent osteoporosis and provide reference for the prevention of osteoporosis by traditional Chinese medicine.
