摘 要:目的建立防风不同炮制品中升麻素苷和5-O-甲基维斯阿米醇苷的含量测定方法并进行比较。方法采用高效液相色谱法(HPLC),使用Hitachi Lachrom-C_(18)色谱柱(150 mm×4.6 mm,5μm),以甲醇-水溶液(40:60)作为流动相,设定检测波长为254 nm、柱温为30℃、流速为1.0 m L/min,进样量3μL。同时,采用快速水分测定仪测定不同炮制品的水分含量。结果升麻素苷和5-O-甲基维斯阿米醇苷分别在14.75~118.00μg/mL和24.00~192.00μg/mL范围内有良好的线性关系。升麻素苷和5-O-甲基维斯阿米醇苷精密度的相对标准偏差(RSD)分别为0.68%、1.13%,重现性的RSD分别为0.64%、1.74%,平均回收率分别为100.80%,95.65%。防风炮制后升麻素苷和5-O-甲基维斯阿米醇苷的含量均有不同程度的下降,虽然微波炮制炒防风的5-O-甲基维斯阿米醇苷含量要低于传统炮制品,但是微波炮制品含量整体高于传统炮制品,炮制后水分含量明显降低,微波炮制品水分含量整体略高于传统炮制品。结论HPLC法快捷、简便、准确、重复性好,可用于防风不同炮制品的质量控制;防风的微波炮制品有效成分含量较高,水分含量略低。Objective To establish a method for simultaneous determination of prim-O-glucosylcimifugin and 5-O-methylvisammioside content in Saposhnikovia divaricata,and compare the contents in different processed products.Methods High performance liquid chromatography(HPLC)was used with the Hitachi Lachrom-C_(18) column(150 mm×4.6 mm,5μm),methanol-water(40:60)were used as the mobile phase,the detection wavelength was set at 254 nm,the column temperature was 30℃,the flow rate was 1.0 mL/min,and the injection volume was 3μL.At the same time,the rapid moisture detector was used to determine the moisture content of different processed products.Results There was a good relationship between prim-O-glucosylcimifugin and 5-O-methylvisammioside in the ranges of 14.75-118.00μg/mL and 24.00-192.00μg/mL,respectively.The relative standard deviation(RSD)of precision of prim-O-glucosylcimifugin and 5-O-methylvisammioside were 0.68%and 1.13%,the RSD of reproducibility were 0.64%and 1.74%,and the average recoveries were 100.80%and 95.65%,respectively.The content of prim-O-glucosylcimifugin and 5-O-methylvisammioside decreased in different degrees after processing.Although the content of 5-O-methylvisammioside in microwave processed fried Saposhnikovia divaricata was lower than that of traditional processed products,the content of microwave processed products was higher than that of traditional processed products,after processing,the moisture content decreased significantly,and the moisture content of microwave processed products was slightly higher than that of traditional processed products.Conclusion The HPLC method is fast,simple,accurate and reproducible,and can be used for the quality control of different processed products of Saposhnikovia divaricata.The microwave processed products of Saposhnikovia divaricata had higher content of active ingredients and slightly lower moisture content.
