摘 要:目的通过建立慢性复合心理应激模型,检测雄性大鼠生殖生理的改变,初步探讨慢性心理应激对雄鼠生殖功能的影响。方法选取健康雄性SD大鼠24只,随机平均分为应激组和对照组。应激组采用慢性复合心理应激模型。通过糖水消耗实验研究慢性心理应激对雄鼠行为学的影响;通过检测体重增长率、睾丸和附睾脏器系数以及Western blot实验检测睾丸雄激素结合蛋白(ABP)和睾丸特异性含溴结构域蛋白(BRDT)表达,来研究慢性心理应激对雄鼠生殖生理指标的影响。结果应激14、21 d,应激组的糖水偏爱系数明显低于对照组(P<0.01)。应激3、7、14、21 d,应激组的体重增长率明显低于对照组(P<0.05)。应激21 d,应激组的睾丸、附睾重量和附睾指数低于对照组(P<0.05)。应激21 d,应激组的精子密度和精子活力低于对照组(P<0.01)。应激21 d,应激组的睾丸ABP和BRDT表达均下调。结论慢性心理应激可引起雄鼠行为学、雄性生理及生殖器官的改变,影响大鼠睾丸蛋白的正常表达模式,从而影响雄鼠的生殖功能。Objective To detect the changes of reproductive physiology in male rats by establishing a chronic compound psychological stress model,and preliminarily explore the effects of chronic psychological stress on reproductive function of male rats.Methods Twenty-four healthy male SD rats were selected and randomly evenly divided into stress group and control group.The stress group used the chronic complex psychological stress model.The effect of chronic psychological stress on the behavior of male rats was studied by sugar water consumption experiment;and the effects of chronic psychological stress on reproductive physiological indexes of male rats were studied by detecting body weight growth rate,organ coefficients of testis and epididymis,and the expression of androgen binding protein(ABP)and bromodomain testis-specific protein(BRDT)by Western blot test.Results At 14,21 d of stress,the sucrose preference coefficient of the stress group was significantly lower than that of the control group(P<0.01).At 3,7,14,21 d of stress,the weight growth rate of the stress group was significantly lower than that of the control group(P<0.05).At 21 d of stress,the weight of testis and epididymis and epididymal index in the stress group were lower than those in the control group(P<0.05).At 21 d of stress,the sperm density and sperm motility of the stress group were lower than those of the control group(P<0.01).At 21 d of stress,the expressions of ABP and BRDT in the testis of the stress group were down-regulated.Conclusion Chronic psychological stress can cause changes in male behavior,male physiology and reproductive organs,affect the normal expression pattern of rat testicular protein,and thus affect the reproductive function of male rats.
