摘  要:由尖孢镰刀菌甜瓜专化型(Fusarium oxysporum f.sp.melonis)引起的枯萎病是一种严重影响甜瓜生产的病害,果胶裂解酶是细胞壁降解酶之一。尖孢镰刀菌在侵染甜瓜的过程中,果胶裂解酶基因是否起到作用,目前相关报道较少。本研究利用同源重组的原理,通过农杆菌(Agrobacterium tumefaciens)介导的遗传转化对甜瓜枯萎病菌果胶裂解酶基因FopLb、FopLe、FopL1和FopL2进行敲除,并对敲除突变体开展表型、致病力、胁迫因子敏感性测定试验。结果显示,突变体菌株ΔFopLb和ΔFopL1菌落小、无色素沉淀,菌丝稀疏且较细,几乎不产生孢子,致病力明显降低。所有突变体菌株在抵抗外源盐胁迫及对纤维素利用能力与野生型菌株基本一致。研究结果表明,FopLb和FopL1在营养生长、无性孢子形成和致病性中起重要作用,ΔFopLb和ΔFopL1菌株严重的生长缺陷可能是导致其致病性减弱的重要原因。The wilt of melon caused by Fusarium oxysporum f.sp.melonis is a disease that seriously affects melon production.Pectin lyase is one of the cell wall degrading enzymes.Whether the pectin lyase gene plays a role in the process of F.oxysporum f.sp.melonis infestation is less reported.This study used the principle of homologous recombination to knock out the FopLb,FopLe,FopL1 and FopL2 via Agrobacterium tumefaciens-mediated genetic transformation,and the knockout mutants were subjected to experiments on phenotypes,pathogenicity,susceptibility to stress factors.Results showed that the mutant strainsΔFopLb andΔFopL1 exhibited smaller colonies without pigment deposition,whose mycelia were sparse and thinner and hardly produced spores,thus significantly reduced pathogenicity.All mutant strains displayed resistance to exogenous salt stress and cellulose utilization capabilities similar to the wild-type strain.The findings suggest that FopLb and FopL1 play crucial roles in nutritional growth,asexual spore formation,and pathogenicity,with the severe growth defects ofΔFopLb andΔFopL1 likely being a major cause of their reduced pathogenicity.
