养精种玉汤对薄型子宫内膜大鼠白细胞介素-1β、核因子κB的影响

摘  要:目的:探讨养精种玉汤对薄型子宫内膜大鼠炎症介质的影响。方法:选取动情周期规律的24只无特定病原体(SPF)级斯泼累格·多雷(SD)雌性大鼠随机分为空白组、中药组与模型组,每组8只。中药组与模型组通过向子宫腔内注射95%无水乙醇建立薄型子宫内膜模型,空白组大鼠不干预。造模后,中药组大鼠予以养精种玉汤灌胃,空白组与模型组以蒸馏水灌胃。灌胃3个动情周期后取子宫组织及动脉血,进行子宫内膜厚度测量,用苏木精-伊红(HE)染色观察大鼠子宫内膜组织形态,用酶联免疫吸附试验法检测血清白细胞介素-1β(IL-1β)、核因子κB(NF-κB)水平,用定量PCR检测子宫组织IL-1βmRNA、NF-κBmRNA相对表达量。结果:与空白组比较,模型组大鼠子宫湿重、子宫脏器指数、内膜厚度、腺体数明显降低(P<0.01),血清IL-1β及NF-κB水平、子宫组织IL-1βmRNA及NF-κB mRNA相对表达量明显升高(P<0.01);与模型组比较,中药组大鼠子宫湿重、子宫脏器指数、内膜厚度、腺体数明显升高(P<0.01),血清IL-1β及NF-κB水平、子宫组织IL-1βmRNA及NF-κB mRNA相对表达量明显降低(P<0.05)。结论:养精种玉汤可增加薄型子宫内膜大鼠的子宫内膜厚度,其作用机制可能与抑制NF-κB、IL-1β有关,即降低炎症介质水平,促进子宫内膜细胞的增殖。Objective:To investigate the effect of Yangjing Zhongyu Decoction on inflammatory factors in rats with thin endometrium.Methods:Twenty-four specific pathogen free(SPF)-grade SD female rats with regular estrous cycles were randomly divided into a blank group,a Chinese medicine group,and a model group,with 8 rats in each group.Rats in the Chinese medicine group and the model group were injected with 95% anhydrous ethanol into the uterine cavity to establish the rat model of thin endometrium,while the blank group received no intervention.After modeling,rats in the Chinese medicine group were administered Yangjing Zhongyu Decoction by gavage,while those in the blank and model groups were given distilled water.After three estrous cycles of treatment,uterine tissue and arterial blood were collected.Uterine endometrial thickness was measured,and hematoxylin-eosin(HE) staining was performed to observe the morphology of the uterine endometrial tissue.Serum levels of interleukin-1β(IL-1β) and nuclear factor κB(NF-κB) were measured using enzyme-linked immunosorbent assay(ELISA),and relative expression levels of IL-1β mRNA and NF-κB mRNA in uterine tissue were determined using quantitative PCR.Results:Compared with the blank group,the model group showed significantly reduced uterine wet weight,uterine organ index,endometrial thickness,and gland number(P<0.01),significantly increased serum levels of IL-1β and NF-κB,and increased relative expression levels of IL-1β mRNA and NF-κB mRNA in uterine tissue(P<0.01).Compared with the model group,the Chinese medicine group exhibited significantly increased uterine wet weight,uterine organ index,endometrial thickness,and gland number(P<0.01),significantly reduced serum levels of IL-1β and NF-κB,and reduced relative expression levels of IL-1β mRNA and NF-κB mRNA in uterine tissue(P<0.05).Conclusion:Yangjing Zhongyu Decoction can increase endometrial thickness in rats with thin endometrium.Its mechanism of action may be related to the inhibition of NF-κB and IL-1β,which
