摘 要:本文以2-(3-醛基-4-羟基苯基)-4-甲基噻唑-5-羟基酸乙酯和2-肼基苯并噻唑为原料合成探针BTMH。在365 nm紫外灯照射下,探李成灿针分子在固态时发射强烈的黄色荧光。荧光光谱实验表明,在乙醇(EtOH)/水(H2O)体系中,随着含水量的增加,溶液的荧光增强且发生红移,表明探针BTMH具有显著的聚集诱导发光(AIE)效应。在包含10μmol/L BTMH的EtOH/H2O(V/V=1/9,pH 7.4)溶液中,加入次氯酸(ClO^(-))后,溶液荧光由黄色变为蓝色,且BTMH-ClO^(-)体系在470 nm处与560 nm处的荧光强度的比值与0~50μmol/L范围内的ClO^(-)浓度呈良好的线性关系,表明探针BTMH能够比率检测ClO^(-)。通过红外(FT-IR)、核磁(1H NMR)、质谱(HRMS)及理论计算(DFT)推断了探针BTMH对ClO^(-)的传感机理。该探针成功用于在PC12细胞中检测内源性及外源性ClO^(-)。In this paper,a probe,BTMH,was synthesized through the reaction of 2-(3-formyl-4-hydroxyphenyl)-4-methylthiazole-5-hydroxy acid ethyl ester and 2-hydrazinobenzothiazole.The probe BTMH in solid state showed strong yellow fluorescence under 365 nm ultraviolet lamp irradiation.With the gradual increase of the volume fraction of water in the ethanol(EtOH)/H2O system,the fluorescence intensity of the solution tended to increase and the wavelength appeared to be red-shifted,which indicated that the solution had obvious aggregationinduced emission(AIE)effect.After the addition of ClO^(-),the fluorescence color of the BTMH solution changed from yellow to blue,and the fluorescence intensity ratio of the BTMH-ClO^(-)system at 470 nm and 560 nm showed a good linear relationship with ClO^(-)concentration in the range of 0-50μmol/L,indicating that probe BTMH could detect ClO^(-)by ratio response.Also,the sensing mechanism of BTMH to ClO^(-)was investigated by 1H nuclear magnetic resonance(1H NMR)spectroscopy,Fourier transform infrared(FT-IR)spectroscopy and high resolution mass spectrometry(HRMS),and was further demonstrated by the functional theory(DFT)method.Further,cells imaging experiments showed that probe BTMH could detect endogenous and exogenous ClO^(-)in PC12 cells successfully.
