川金丝猴源肺炎克雷伯菌的分离鉴定及耐药性

摘　　要：从急性死亡川金丝猴(Rhinopithecus oxellana)的肺脏、心脏、肝脏等病料组织中分离到一株革兰氏阴性杆菌。通过对该菌株进行培养和形态观察,发现其特征与肺炎克雷伯菌相符。利用PCR技术对该菌株的16S rRNA基因进行扩增,得到一个长约1433 bp的特异性DNA片段,并将其序列通过NCBI数据库进行BLAST分析,该序列与人源肺炎克雷伯菌肺炎亚种01A030株(参考序列NR_134062.1)核苷酸序列相似性达到99.7%。通过对10种肺炎克雷伯菌致病性相关的毒力基因进行PCR扩增,其中有8种能够被检测到,即rmpA、fimH、wabG、uge、kfu、aerobactin、ureA和khe;根据上述结果对该菌株进行拉丝试验,结果为阳性,确证成功分离一株高毒力肺炎克雷伯菌(hypervirulent Klebsiella pneumonia,hvKP)。药敏试验结果显示,该hvKP分离株对第一代头孢和第三代头孢药物表现为不同程度耐药,对林可酰胺类克林霉素、林可霉素均完全耐药;对沙星类药物,如氧氟沙星、恩诺沙星及环丙沙星等表现出敏感性。A Gram-negative bacterium was isolated from the lung,heart,liver and other diseased tissues of acute dead golden monkey(Rhinopithecus oxellana).Culture and morphological observation of this bacterium were found to coincide with K.pneumoniae characteristics.The 16S rRNA gene of this strain was amplified by PCR with a specific DNA fragment approximately 1433 base pairs long and BLAST using the NCBI database that showed 99.7%nucleotide sequence similarity to the human K.pneumoniae substrain 01A030(reference sequence NR_134062.1).Ten virulence genes related to the pathogenicity of K.pneumoniae were selected,and eight virulence genes(rmpA,fimH,waG,uge,kfu,aerobactin,ureA,khe)were tested by PCR,combined with positive strain drawing test,one hvKP was successfully isolated.The results showed that the hvKP isolate was resistant to the first and third generation cephalosporins,completely resistant to lincoamide clindamycin and sensitive to afloxacin,such as ofloxacin,enrofloxacin and ciprofloxacin.
