摘 要:为探究环状RNA circ_1764在鸡脂肪细胞分化中的重要作用,试验通过PCR扩增和RNase酶消化鉴定鸡circ_1764的存在特性,采用qRT-PCR技术检测circ_1764在鸡组织中的表达情况,利用Miranda、miRDB和Target Scan Human等网站预测鸡circ_1764的调控网络,使用DAVID软件可视化预测靶基因的GO和KEGG富集通路,最后通过转染过表达circ_1764载体,验证生物信息学筛选出的调控网络的准确性。结果显示:鸡体内存在circ_1764,并主要在细胞质中表达,在鸡心脏、肝脏、胸腺和脂肪组织中高表达;circ_1764的预测靶miRNA主要为miR-7441-3p、miR-1656、miR-6578-3p、miR-7452-5p、miR-6645-5p、miR-6563-3p、miR-6607-5p和miR-6581-5p,并且circ_1764在DF-1细胞中过表达可以显著降低上述miRNA的表达;circ_1764靶基因主要集中在代谢、泛素介导的蛋白水解以及MAPK通路中。本研究验证了鸡circ_1764的存在,通过生物信息学预测了鸡circ_1764的调控网络,初步探究circ_1764通过吸附miR-1656、miR-6645-5p等miRNA调控靶基因进而影响鸡脂肪细胞分化的分子机制,为进一步阐明circ_1764调控鸡脂肪细胞分化的机理奠定了基础。To investigate the role of circular RNA circ_1764 in chicken adipocyte differentiation,the biological features chicken circ_1764 were characterized by PCR amplification and RNase digestion.Additionally,the expression of circ_1764 in chicken tissues was quantified using qRT-PCR technology.The regulatory network of chicken circ_1764 was predicted using on-line tools such as Miranda,miRDB,and TargetScan.Subsequently,the DAVID software was used to visualize the GO and KEGG enrichment pathways of predicted target genes,and the accuracy of the regulatory network was verified by the overexpression circ_1764 vector through transfection.The results demonstrated the existence of circ_1764 in chicken,with predominant expression in the cytoplasm,which was highly expressed in the heart,liver,thymus and adipose tissue.Furthermore,the predicted targeted miR-NAs of circ_1764 included miR-7441-3p,miR-1656,miR-6578-3p,miR-7452-5p,miR-6645-5p,miR-6563-3p,miR-6607-5p and miR-6581-5p.Moreover,the expression of these miRNAs was significantly decreased due to overexpression of circ_1764 in DF-1 cells.The target genes of circ_1764 showed significant enrichment in the metabolism,ubiquitin-mediated proteolysis,and MAPK pathways.This study confirmed the presence of chicken circ_1764,predicted its regulatory network using bioinformatics,and preliminarily investigated the role of circ_1764 in regulating target genes by absorbing miR-1656,miR-6645-5p,and other miRNAs,thereby contributing to the regulation of chicken adipocyte differentiation.The study provided a ba-sis for further clarifying the mechanism by which circ_1764 regulates chicken adipocyte differentiation.
