科尔沁牛不同组织脂肪酸含量与SCD和SREBP-1基因表达相关分析

摘  要:文章旨在研究科尔沁牛不同组织脂肪酸含量及SCD、SREBP-1基因表达量和参与脂肪酸代谢的调控机制。试验采用气相色谱-质谱联用法和实时荧光定量PCR方法分别检测科尔沁牛的肾周脂肪、肠周脂肪、肩肌和背最长肌等4个不同组织的脂肪酸含量及SCD和SREBP-1基因的表达量。结果表明:(1)SFA中,肾周脂肪C14:0、C16:0、C18:0含量显著高于背最长肌(P<0.05)。MUFA中,背最长肌C16:1和总MUFA含量高于肾周脂肪,但无显著差异(P>0.05)。背最长肌的各PUFA、总PUFA和UFA含量均显著高于肾周脂肪(P<0.05)。(2)SCD基因在科尔沁牛背最长肌中的表达量高于肾周脂肪组织,与SREBP-1基因的表达趋势相似,但无显著差异(P>0.05)。SREBP-1基因在科尔沁牛背最长肌中的表达量显著高于肾周脂肪组织(P<0.05)。以上结果表明,科尔沁牛背最长肌的营养价值与肉质较肾周脂肪更高、更有利于健康。同时,SREBP-1基因在参与SCD基因的脂肪沉积调控过程中,协同调节肌内脂肪代谢,进而影响牛肉脂肪酸组成及风味。This article aims to study the differences in fatty acid content,as well as the expression levels of SCD and SREBP-1 genes involved in fatty acid metabolism regulation in different tissues of the Horqin cattle.Gas chromatography-mass spectrometry and real-time fluorescence quantitative PCR were used to detect the fatty acid content in two different tissues of Horqin cattle,namely the perirenal fat,perienteric fat,shoulder muscle,and longissimus dorsi,as well as the expression levels of the SCD and SREBP-1 genes.The results show that:(1)In saturated fatty acids(SFA),the content of C14:0,C16:0,and C18:0 in perirenal fat is significantly higher than that in the longissimus dorsi(P<0.05).In monounsaturated fatty acids(MUFA),the content of C16:1 and total MUFA in the longissimus dorsi is higher than in perirenal fat,but no significant difference(P>0.05).The content of each polyunsaturated fatty acid(PUFA),total PUFA,and unsaturated fatty acids(UFA)in the longissimus dorsi are all significantly higher than in perirenal fat(P<0.05).(2)The expression level of the SCD gene in the longissimus dorsi of Horqin cattle is higher than in perirenal fat tissue,showing a similar trend to the expression of the SREBP-1 gene,but no significant difference(P>0.05).The expression level of the SREBP-1 gene in the longissimus dorsi of Horqin cattle is significantly higher than in perirenal fat tissue(P<0.05).These results indicate that the nutritional value and meat quality of the longissimus dorsi of Horqin cattle are higher and more beneficial to health than perirenal fat.At the same time,the SREBP-1 gene is involved in the regulation of fatty acid deposition along with the SCD gene,co-regulating intramuscular fat metabolism,thereby influencing the composition and flavor of beef fatty acids.
