摘 要:为探讨附子理中丸通过NLRP-3炎症小体途径的抗炎止泻作用机制,将Wistar大鼠分为对照组、模型组、小檗碱组、IKK-β抑制剂组(简称抑制剂组)以及附子理中丸低、中、高剂量组(分别为10、20、40 g·kg-1)。模型组、小檗碱组、抑制剂组及低、中、高剂量组经腹腔注射伊立替康(irinotecan, CPT-11)溶液,建立CPT-11诱导的大鼠腹泻模型。建模后,小檗碱组、抑制剂组及低、中、高剂量组灌胃相应的药物,模型组、对照组给予等容量的蒸馏水,连续灌胃30 d后取材检测相关指标。结果表明:与对照组相比,模型组结肠组织病理变化及其超微结构损伤严重;IL-1β、IL-18、TNF-α水平及NLRP-3、Caspase-1、ASC、GSDMD的mRNA相对表达量和NLRP-3、Caspase-1、ASC蛋白相对表达量均显著升高。与模型组相比,各给药组均可不同程度地改善上述指标和病理变化。这一研究提示附子理中丸可抑制NLRP-3炎症小体的激活,从而改善肠道炎症与腹泻。To investigate the anti-inflammatory and antidiarrheal effects of Fuzi Lizhong pill and to study its mechanisms through NLRP-3inflammasomes.Wistar rats were divided into control group,model group,berberine group,IKK-βinhibitor group,Fuzi Lizhong pill low,middle and high dose groups(10,20,40g·kg-1 respectively).A rat model of irinotecan-induced diarrhea was established in the model group,berberine group,IKK-βinhibitor group,Fuzi Lizhong pill low,middle and high dose groups.After successful modeling,except for the model group,each group was given the corresponding drug for gastric gavage,and the control group was given distilled water of equal volume,and the gastric gavage was continuously gavaged for 30days and then to detect relevant indexes.The results showed that in the model group the histopathological changes of the colon in the model group and its ultrastructural damage were serious,moreover,the levels of IL-1β,IL-18and TNF-α,the relative expressions of mRNA of NLRP-3,Caspase-1,ASC,GSDMD and the relative expressions of NLRP-3,Caspase-1and ASC proteins all significantly increased compared to the control group.However,berberine group,IKK-βinhibitor group,Fuzi Lizhong pill low,middle and high dose groups can improve above indexes in different degrees compared to the model group.It is supposed that Fuzi Lizhong pill can alleviate inflammation and diarrhea by inhibiting the activation of NLRP-3inflammasomes.
