摘 要:目的建立专属有效的朝药材肾炎草薄层色谱鉴别方法,加快其质量标准制定进程。方法基于固相萃取技术制备对照药材及供试品溶液,点样量为5μl,展开剂为三氯甲烷-甲醇-水(45∶10∶1),显色剂为10%硫酸乙醇溶液,显色条件为105℃加热至斑点显色清晰,日光下检视。结果所得薄层色谱图斑点清晰,主斑点比移值适中;从低温、低湿、高湿,不同品牌硅胶G板、不同品牌C_(18)固相萃取小柱及不同来源药材等多方面考察,验证方法耐用性良好。结论本实验建立了肾炎草薄层色谱鉴别方法,层次清晰、稳定可靠、专属性及有效性较强,为肾炎草药材的质量控制提供有效依据。Objective To establish a stable,effective and specific TLC identification method for Korean medicine Me-landrium firmum Rohrb.and to accelerate the quality standard setting process of Me-landrium firmum Rohrb.for promoting the development of ethnic medicine.Methods The test solution and reference solution were prepared based on SPE technology.The sample size was 5μl.The developing agent was trichloromethane-methanol-water(45∶10∶1),while the chromogenic reagent was 10%sulfuric acid and ethanol solution.The plate was heated at 105℃until the spots were clear and the TLC plate was inspected under sunlight.Results In the TLC figures,the spots were clear and the Rf values of the main spots were moderate.In terms of several factors such as low temperature environment,low humidity environment,high humidity environment and different brands of thin layer plates and C18 SPE columns,the method had well durability.Conclusion This experiment establishes a TLC identification method for Me-landrium firmum Rohrb.,which is stable,reliable,specific and effective.And this method provides an effective approach for controlling the quality of medicinal materials.
