摘 要:目的验证阳离子脂质体介导神经营养因子3(Neurotrophin-3,NT3)基因转染对噪声致豚鼠耳聋的保护作用。方法选取广西医科大学动物实验中心提供的60只健康花色黑目豚鼠为研究对象,按随机数字表法分为5组,每组12只(实验过程中部分动物死亡,实际实验数据按每组10只统计)。Ⅰ组未受噪声影响,Ⅱ组注入人工外淋巴液未受噪声刺激,Ⅲ组注入人工外淋巴液后受噪声刺激,Ⅳ组在注入空质粒后受噪声刺激,V组在注入含NT3基因的质粒后受噪声刺激。噪声为4 kHz的稳态白噪声,强度100 dB,8 h/d,连续10 d,休息5 d,再通过听性脑干反应(Auditory Brain-Stem Response,ABR)测试和免疫组织化学染色来评估听阈变化和耳蜗螺旋神经节细胞中NT3蛋白的表达情况。结果V组ABR阈值高于Ⅰ、Ⅱ组,但低于Ⅲ、Ⅳ组,差异有统计学意义(P均<0.05)。耳蜗螺旋神经节细胞处NT3蛋白表达量以V组较其余各组更高,差异有统计学意义(P均<0.05)。结论NT3基因转染后阈值增幅减小,听功能受损有一定程度减轻,对噪声致豚鼠耳聋具有一定程度的保护作用。Objective To verify the protective effect of cationic liposomes mediated neurotrophin-3(NT3) gene transfection on noise induced hearing loss in guinea pigs.Methods 60 healthy black eye guinea pigs of different colors were selected as the study objects from the Animal Experiment Center of Guangxi Medical University and were randomly divided into 5 groups by random number table method,with 12 animals in each group(some animals died during the experiment,and the actual experimental data was calculated based on 10 animals in each group).Group I was not affected by noise,Group Ⅱ was not stimulated by noise after injection of artificial external lymph,Group Ⅲ was stimulated by noise after injection of artificial external lymph,Group Ⅳ was stimulated by noise after injection of empty plasmid,and Group V was stimulated by noise after injection of plasmid containing NT3 gene.Steady state white noise with a noise intensity of 4 kHz,100 dB,8 h/d,continuous for 10 days,resting for 5 days.Then,auditory brainstem response(ABR) testing and immunohistochemical staining were used to evaluate changes in hearing threshold and expression of NT3 protein in cochlear spiral ganglion cells.Results The ABR threshold range of Group V was higher than Group I and Group Ⅱ,but lower than Group Ⅲ and Group Ⅳ,and the differences were statistically significant(all P<0.05).The expression level of NT3 protein in the spiral ganglion cells of the cochlea in Group V was higher than those in the other groups,and the differences were statistically significant(all P<0.05).Conclusion After NT3 gene transfection,the threshold increase decreases,and the impairment of hearing function is alleviated to a certain extent,which has a certain degree of protective effect on noise induced deafness in guinea pigs.
