白花蛇舌草鲜品水提液和醇提液的抗炎作用研究

摘　　要：目的:比较白花蛇舌草鲜品水提液和醇提液体内外抗炎作用的差异,为进一步的临床研究提供理论依据。方法:用水或75%乙醇对白花蛇舌草鲜品进行回流提取,并对提取液进行浓缩和冷冻干燥,得到提取物粉末;将提取物粉末配置相应的生药溶液,观察其对炎症模型的小鼠巨噬细胞(RAW264.7)和小鼠的抗炎作用,并比较水提液和醇提液在其中的作用差异。结果:白花蛇舌草鲜品水提液和醇提液均可抑制脂多糖致炎的RAW264.7的活力,并且总体上水提液对致炎的RAW264.7中白介素-6、白介素-1β、肿瘤坏死因子-α等炎症因子的抑制率优于醇提液;此外,白花蛇舌草鲜品水提液和醇提液均能减轻炎症模型小鼠耳肿胀和减少扭体次数,且水提液的整体效果优于醇提液。结论:白花蛇舌草水提液和醇提液的抗炎作用有差异,水提液综合抗炎效果优于醇提液,这可能与其成分差异有关。Objective:To compare the differences in the anti-inflammatory effects in vivo and in vitro of fresh Hedyotis diffusa water extract and alcohol extract,so as to lay a theoretical foundation for further clinical studies.Methods:Fresh Hedyotis diffusa was subjected to refluxing extraction with water or 75%ethanol,and the extract was concentrated and freeze-dried to obtain the extract powder.The extract powder was prepared into the corresponding herbal medicine solution,then the anti-inflammatory effects on mouse macrophages(RAW264.7)and mice in an inflammatory model were observed,and the differences between the water extract and alcohol extract were compared.Results:Both the water extract and the alcohol extract of fresh Hedyotis diffusa inhibited the viability of lipopolysaccharide-induced RAW264.7,and the water extract was generally superior to the alcohol extract in terms of the inhibition rate of inflammatory factors such as interleukin-6,interleukin-1β,and tumor necrosis factor-α.In addition,both the water extract and the alcohol extract of fresh Hedyotis diffusa reduced ear swelling and writhing times in inflammatory model mice.The overall effect of the water extract was superior to the alcohol extract.Conclusion:The anti-inflammatory effects of the water extract and the alcohol extract of Hedyotis diffusa are different,and the former was superior to the latter,which may be correlated with the difference in their components.
