摘  要:为探明西花蓟马(Frankliniella occidentalis)离子型受体(ionotropic receptor,IRs)的功能、作用机制及其在防治中的应用,本研究通过DNAMAN软件进行序列分析,采用BLAST进行同源性比较,采用MEGA 6的邻接法(neighbor-joining)构建系统进化树,对其序列特征及系统进化情况进行了研究。结果表明,成功克隆了一个西花蓟马离子型受体基因,命名为FoccIR01(GenBank登录号:PP314350);该基因序列cDNA全长3 103 bp,其中离子型受体蛋白基因读码框全长2 415 bp (ORF),编码805个氨基酸,3′端非编码区217 bp,5′端非编码区468 bp;预测成熟蛋白分子量约为90010.90 Da,等电点为6.95;对FoccIR01序列进行同源相似性搜索,结果显示属于鳞翅目昆虫的最多,占总量的60%;进化系统分析表明,FoccIR01与西花蓟马的FoccIR40aX3 (GenBank登录号:XP_052122665.1)、FoccIR40a (GenBank登录号:KAE8747820.1)、FoccIR40aX1(GenBank登录号:XP_052122663.1)和FoccIR40aX2(GenBank登录号:XP_052122664.1)亲缘关系最近,其次是棕榈蓟马TpalIR40a(GenBank登录号:XP_034250854.1)、豆大蓟马MusiHP(GenBank登录号:KAJ1529788.1)以及长叶异痣蟌IeleIR40a(GenBank登录号:XP_046401562.1),表明FoccIR01与这些昆虫的亲缘关系相对较近。本研究成功克隆了一个西花蓟马离子型受体基因FoccIR01,明确了该蛋白基因的核苷酸、氨基酸序列特征,为进一步研究该蛋白基因在西花蓟马行为活动中发挥的具体作用奠定了基础。In order to explore the function and mechanism of IRs and their application in the prevention and control of Frankliniella occidentalis,sequence analysis was performed using DNAMAN software,homology comparison was conducted with BLAST,and a phylogenetic tree was constructed using MEGA 6's neighbor-joining method. Its sequence characteristics and phylogenetic status were studied in this paper.An ionic receptors gene named FoccIR01(GenBank accession number:PP314350) was successfully cloned in Frankliniella occidentalis. The full length of FoccIR01 cDNA is 3 103 bp,contains a 2 415 bp open reading frame(ORF) encoding a putative protein of 805 amino acids with a molecular mass of 90010.90Da and an isoelectric point of 6.95. The non-coding regions of the 3' end and 5' end are 217 bp and468 bp,respectively. The results from the homology similarity search showed that insects belonging to the order Lepidoptera were the most,accounting for 60% of the total. Evolutionary system analysis showed that FoccIR01 has the closest phylogenetic relationship with FoccIR40aX3(GenBank accession number:XP_052122665.1),FoccIR40a(GenBank accession number:KAE8747820.1),FoccIR40aX1(GenBank accession number:XP_052122663.1) and FoccIR40aX2(GenBank accession number:XP_052122664.1) from Frankliniella occidentalis,followed by TpalIR40a(GenBank accession number:XP_ 034250854.1) in Thrips palmi,Musi HP(GenBank accession number:KAJ1529788.1) in Megalurothrips usitatus and IeleIR40a(GenBank accession number:XP_046401562.1) in Ischnura elegans,which indicated that FoccIR01 is relatively closely related to them. Clarified the nucleotide and amino acid sequence characteristics of FoccIR01,laid the foundation for further research on the specific role of this protein gene in the behavioral activities of Frankliniella occidentalis.
