摘  要:为建立窄穗组常见入侵杂草的DNA条形码鉴定方法,选取核基因组的ITS2、ITS,叶绿体基因组的mat K、ndh F、trn T-trn L、rpl32-trn L、rpo B-trn C和trn C-pet N等8个片段进行筛选。综合进化树、种间差异,barcoding gap等多项指标,结果表明trn T-trn L、rpl32-trn L、trn C-pet N和rpo B-trn C序列鉴定效果相对较好,ITS2、ndh F、ITS和mat K序列分辨能力较差。基于叶绿体基因组的trn T-trn L和rpl32-trn L序列,在扩增测序的成功率、种间变异、物种鉴定能力、聚类分析等均表现较好,虽然在一些种内存在较高的种内变异,在形态鉴定的基础上可作为窄穗组DNA条形码鉴定的推荐片段。We tested the applicability of eight DNA regions(ITS2、ITS、mat K,ndh F,trn T-trn L,rpl32-trn L,rpo B-trn C and trn C-pet N) as the barcodes for identifying invasive weeds in Bromus Sect. Genea. The results showed that trn T-trn L,rpl32-trn L,trn C-pet N and rpo B-trn C sequences were better identified,as ITS2,ndh F,ITS and mat K sequences were poorly resolved. The chloroplast genome-based trn T-trn L and rpl32-trn L sequences performed well in terms of amplification and sequencing success,interspecific variation,species identification ability,and cluster analysis,despite high intraspecific variation in some species.Based on morphological identification,these sequences can be recommended as DNA barcode fragments for Bromus Sect. Genea.
