摘　　要：建立白花蛇舌草注射液芦丁、白麻苷及总黄酮高效液相色谱含量测定方法。使用HPLC法对白花蛇舌草注射液中芦丁、白麻苷及总黄酮含量进行测定,色谱柱为InertSustainSwift C18(4.6 mm×250mm,5μm)柱;流动相为乙腈(A)-0.1%醋酸水溶液(B),采用梯度洗脱,柱温30℃;体积流量1.0 mL/min;检测波长369 nm。芦丁进样量在0.201 6~3.225 6μg范围内线性关系良好(R^(2)=0.999 2),平均回收率(n=6)为97.3%;白麻苷在0.196 0~3.136 0μg范围内线性关系良好(R^(2)=0.999 3)平均回收率(n=6)为97.3%;总黄酮平均回收率(n=6)为98.9%,RSD为1.46%。白花蛇舌草注射液中芦丁的平均含量(n=3)为0.08 mg/mL;白麻苷的平均含量(n=3)为0.20 mg/mL总黄酮含量为1.26 mg/mL。本研究方法结果准确、方便快速,为白花蛇舌草注射液中芦丁、白麻苷及总黄酮含量测定提供了新方法。To establish a HPLC method for the determination of rutin,quercetin-3-O-sophoroside and total flavonoids in Scleromitrion diffusum(wild.)R.J.Wang injection.The content of rutin,quercetin-3-O-sophoroside and total flavonoids in Scleromitrion diffusum(wild.)R.J.Wang injection were determined by HPLC on InertSustainSwift C18(4.6mm 250 mm,5 m)column,acetonitrile(A)-0.1%acetic acid aqueous solution(B)with gradient elution,column temperature 30℃,volume flow rate of 1.0 mL/min,detection wavelength 369 nm.The injection amount of rutin had a good linear relationship(R^(2)=0.9992)and an average recovery rate(n=6)of 97.3%in the range of 0.2016~3.2256 g,a linear relationship of quercetin-3-O-sophoroside in the range of 0.1960~3.1360 g(R^(2)=0.9993),an average recovery rate of 97.3%,an average recovery rate of total flavonoids(n=6)of 98.9%,and an RSD of 1.46%.The average content of rutin in Scleromitrion diffusum(wild.)R.J.Wang injection(n=3)was 0.08 mg/mL.The mean content of quercetin-3-O-sophoroside(n=3)was 0.20 mg/mL and the total flavonoid content was 1.26 mg/mL.The method is accurate,convenient and rapid in this study and provides a newmethod for the determination of rutin,quercetin-3-O-sophoroside and total flavonoids in Scleromitrion diffusum(wild.)R.J.Wang injection.
