摘  要:通过云南独蒜兰种子无菌萌发获得原球茎;原球茎经过增殖长叶、生根培养获得组培苗;然后在温度为15、20、25、30、35℃下进行分别培养,光照时间12 h/d,光照强度1500 lx。挑选出30℃(T1)和35℃(T2)的组培苗转入变温条件(白天温度25℃、光照时长12 h,晚上温度20℃、光照时长12 h)的培养箱内进行培养,对照(CK)培养条件为温度25℃(光照时长24 h),光照强度均为1500 lx。定期观察云南独蒜兰叶片数量和长度、假鳞茎数量和大小、须根数。结果表明:假鳞茎数量、大小在35℃培养条件下最高,15℃培养条件下最低;叶片长度在25℃培养条件下最高,35℃培养条件下最低;叶片数量在25℃培养条件下最高,35℃培养条件下最低;须根数在35℃培养条件下最高,20℃培养条件下最低。通过变温继续培养后,假鳞茎数量、大小和须根数T2最多,叶片数量和叶片长度CK组最多。通过炼苗对比,传统云南独蒜兰组培苗炼苗成活率为83.3%,成苗率为72.4%,变温处理后云南独蒜兰组培苗炼苗成活率达到94.6%,成苗率达到91.2%。Protocorm was obtained by aseptic germination of Pleione yunnanensis seeds.Tissue culture seedlings were obtained from protocorm by proliferation,long leaves and rooting culture.They were cultured at 15℃,20℃,25℃,30℃and 35℃respectively,and the illumination time was 12 h/d and the illumination intensity was 1500 lx.Select tissue culture seedlings at 30℃(T1)and 35℃(T2)and transfer them to a temperature changing incubator(with a daytime temperature of 25℃and a light duration of 12 hours,and a nighttime temperature of 20℃and a light duration of 12 hours)for cultivation.The culture conditions of control group(CK)were light time 12 h/d,light intensity 1500 lx,temperature 20℃(night)and 25℃(day).The number and length of leaves,the number and size of pseudokirin,and the number of fibrous roots were observed regularly.The result showed that the number and size of pseudokirin were the highest under 35℃culture conditions and lowest under 15℃culture conditions.The leaf length was the highest in 25℃culture,and the lowest in 35℃culture conditions.The number of leaves in 25℃culture conditions was the highest,and the lowest in 35℃culture conditions.The number of fibrous roots was the highest at 35℃,and the lowest at 20℃culture conditions.After continuous culture at variable temperature,the number and size of the pseudonymous,and number of fibrous roots were the highest in the T2 group,while the number and length of leaves were the highest in the CK group.By comparison of hardening-seedlings,the survival rate of tissue culture hardening seedlings of traditional Pleione yunnanensis was 83.3%,and the seedling formation rate was 72.4%,the survival rate of tissue culture seedlings of Pleione yunnanensis after variable temperature treatment was 94.6%,and the seedling formation rate was 91.2%.
