摘　　要：目的探讨血管内皮生长因子(VEGF)预处理对大鼠肝脏缺血-再灌注损伤的保护作用。方法14只健康Wistar大鼠均分为两组。建立肝脏缺血-再灌注损伤模型前30min,A组和C组经腹腔肠系膜静脉分别注射100μg/kg的VEGF0.5mL和PBS0.5mL。1周后观察两组大鼠存活情况,采用ELISA法和Westernblot法分别检测两组血清VEGF水平和肝脏组织中VEGF蛋白表达,采用免疫组化染色和HE染色分别观察两组肝脏组织中VEGF蛋白表达和肝脏组织形态学变化,比较两组血清ALT、乳酸脱氢酶(LDH)及胆红素水平。结果1周后,A组存活5只,C组存活6只,两组存活比例相仿(P>0.05)。与C组比较,A组血清VEGF水平和肝脏组织中VEGF蛋白表达均升高(P<0.01)。A组肝脏组织中VEGF蛋白多呈棕黄色阳性表达,肝脏组织炎症减轻,水肿及细胞变性程度减轻。A组血清ALT、LDH及胆红素水平均低于C组(P<0.05)。结论VEGF预处理对大鼠肝脏缺血-再灌注损伤具有保护作用。Objective To investigate the protective effect of vascular endothelial growth factor(VEGF)pretreatment on hepatic ischemia-reperfusion injury in rats.Methods Fourteen healthy Wistar rats were equally divided into two groups and were injected with 100μg/kg of VEGF 0.5 mL(group A)and PBS 0.5 mL(group C)through peritoneal mesenteric vein 30 minutes before establishment of hepatic ischemia-reperfusion injury model.After one week,the survival conditions of the rats were observed,serum VEGF level and VEGF protein expression of liver tissues were detected by ELISA and Western blot,respectively.VEGF protein expression and liver histopathological changes were observed by immunohistochemical staining and HE staining,respectively.Serum levels of ALT,lactate dehydrogenase(LDH)and bilirubin were compared between the two groups.Results After one week,5 rats were survived in group A and 6 rats were survived in group C.Survival rate of group A was similar to that of group C(P>0.05).Compared with group C,serum VEGF level and VEGF protein expression of liver tissues were increased in group A(P<0.01).The expression of VEGF protein of liver tissues was mostly brown and yellow positive expression,and the inflammation,edema and cell degeneration of liver tissues were alleviated in group A.Serum levels of ALT,LDH and bilirubin in group A were lower than those in group C(P<0.05).Conclusion VEGF pretreatment has protective effect on the liver ischemia-reperfusion injury in rats.
