摘 要:银屑病主要是由免疫介导、遗传与环境共同作用的难治性疾病。肿瘤坏死因子-α(TNF-α)相关生物制剂为银屑病治疗带来里程碑式进步,然而,抗TNF-α疗法存在不良答应,其局限性可能与TNF-α不同受体激活后发挥的生物学功能不同有关。肿瘤坏死因子受体2(TNFR2)是TNF-α的关键受体之一,在与TNF-α结合后,可激活核因子-κB(NF-κB)、磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/Akt)、丝裂原活化蛋白激酶(MAPK)、转录激活子3(STAT3)等多个信号通路,参与炎症、表皮稳态、细胞凋亡、细胞增殖、细胞自噬等生物学过程的调控,提示TNFR2与银屑病的发生、发展关系密切。既往研究往往忽视TNFR2在抗TNF-α疗法中的作用,因此,该文综述了TNFR2的结构、信号转导通路、在疾病中的研究进展及其与银屑病的关系,为探索银屑病的发病机制和治疗提供新的参考。Psoriasis is a refractory disease mainly co-acted by immune,genetic and environment.Tumor necrosis factorα(TNF-α)-related biologics have brought the landmark advances in the treatment of psoriasis;however,the anti-TNF-αtherapy has the adverse response,its limitation may be related to the different biological functions exerted by activation of TNF-αdifferent receptors.Tumor necrosis factor receptor 2(TNFR2)is one of the key receptors for TNF-α,and after binding to TNF-α,it can activate multiple signaling pathways such as NF-κB,PI3K/Akt,MAPK,STAT3,etc.,which are involved in the regulation of inflammation,epidermal homeostasis,cellular apoptosis,cellular proliferation,cellular autophagy and other biological processes.It is suggested that TNFR2 is closely related to the occurrence and development of psoriasis.Previous studies have often overlooked the role of TNFR2 in anti-TNF-αtherapies;therefore,this article reviews the structure and signaling pathways of TNFR2,research advances in the disease,and its relationship with psoriasis to provide new references for exploring the pathogenesis and treatment of psoriasis.
