摘 要:进一步明确TLR4及其相关免疫因子在禽类疫苗免疫过程中的表达变化。采用荧光定量RT-PCR方法和免疫组织化学技术,在注射H5亚型禽流感疫苗24 h后,对三花鹅TLR4及相关免疫因子在不同组织中的表达情况进行检测并分析。在H5亚型禽流感疫苗诱导下,实验组TLR4 mRNA在肝脏和骨髓中显著高于对照组(P<0.05),在脾脏、法氏囊以及胸腺中极显著高于对照组(P<0.01);IL-1β、IFN-α以及IFN-γ在肝脏和胸腺中极显著高于对照组(P<0.01),IL-6和IFN-α在脾脏中与对照组相比为极显著(P<0.01),但IL-1β在脾脏和法氏囊中只是显著高于对照组的水平(P<0.05)。免疫组织化学检测结果显示,TLR4在脾脏和法氏囊中与对照组相比极显著(P<0.01),在肝脏中为显著差异(P<0.05),与转录水平一致。在H5亚型禽流感疫苗诱导下,三花鹅体内能够激活TLR4及其相关免疫因子,但在不同组织中表达具有一定差异性。In order to make a profound study on expressive characteristics of TLR4 and associated immune factors during poultry vaccination.We used the Sanhua goose as experimental animal to analyze TLR4 and associated immune factors level in different tissues by real⁃time PCR method and immunohistochemistry technique on 24h after injection of H5 subtype avian influenza vaccine(AIV).The results showed that TLR4 mRNA was higher than the control group in liver and bone marrow(P<0.05),It was significantly higher in spleen,bursa of Fabricius and thymus than that in control group(P<0.01).IL⁃1β,IFN⁃αand IFN⁃γmRNA level are significantly higher than those of the control group in liver and thymus(P<0.01),IL⁃6 and IFN⁃αwith significant differences in spleen(P<0.01),but IL⁃1βmRNA level,were higher than control group in spleen and bursa(P<0.05).The immunohistochemical test indicated that TLR4 protein levels was significant higher than control group(P<0.01),and higher than in liver(P<0.05),which was consistent with the transcriptional level.It suggested that TLR4 and associated immune factors might play roles by injection H5 subtype AIV.
