摘 要:目的:基于气相色谱-质谱法(GC-MS)和网络药理学对柴胡Bupleurum chinense DC.、狭叶柴胡B.scorzonerifolium Willd.、窄竹叶柴胡B.marginatum var.stenophyllum(Wolff)Shan et Y.Li、竹叶柴胡B.marginatum Wall.ex DC.和黑柴胡B.smithii Wolff干燥根的挥发油成分及潜在药理活性进行比较研究。方法:采用水蒸气蒸馏法提取5种柴胡根中的挥发油,使用GC-MS对挥发油成分进行定性分析,构建成分-靶点网络并进行京都基因与基因组百科全书富集分析。结果:5种柴胡挥发油中化合物组成存在一定差异,共鉴定出其中的55个化合物;网络药理学研究结果显示,5种柴胡挥发油具有相似的作用靶点,如核转录因子-κB亚基1(NFKB1)、磷酸肌醇3-激酶调节亚基1(PIK3R1)、磷脂酰肌醇-4,5-二磷酸3-激酶催化亚基δ(PIK3CD)、诱导型一氧化氮合酶(NOS2)等,其中共有核心靶点作用于神经活性配体-受体相互作用、中性粒细胞胞外诱捕网形成作用、鞘脂信号通路及小细胞肺癌等信号通路。结论:5种柴胡挥发油在抗感染、抗炎、保护神经及抗肿瘤方面具有潜在的相互替代作用。Objective:To compare the volatile oil components and potential pharmacological activities of dried roots of Bupleurum chinense DC.,B.scorzonerifolium Willd.,B.marginatum var.stenophyllum(Wolff)Shan et Y.Li,B.marginatum Wall.ex DC.,and B.smithii Wolff based on gas chromatography-mass spectrometry(GC-MS)and network pharmacology.Methods:The volatile oils from the roots of these five Bupleurum species were extracted by steam distillation.GC-MS was employed for qualitative analysis of the volatile oil components.A component-target network was constructed,and enrichment analysis was performed using the Kyoto Encyclopedia of Genes and Genomes(KEGG).Results:There were differences in the composition of volatile oil compounds among the five Bupleurum species,with a total of 55 compounds identified.Network pharmacology analysis indicated that the volatile oils of these five Bupleurum species shared similar target proteins,such as NFKB1,PIK3R1,PIK3CD,and NOS2.These core targets were involved in pathways including neuroactive ligand-receptor interaction,neutrophil extracellular trap formation,sphingolipid signaling pathway,and small cell lung cancer signaling pathways.Conclusion:The volatile oils of these five Bupleurum species potentially exhibit mutual substitution effects in anti-infection,anti-inflammatory,neuroprotective,and anti-tumor activities.
