摘  要:目的:探讨忍冬藤提取物对骨质疏松性骨折大鼠模型的影响及其机制。方法:将大鼠随机分为对照组,模型组,忍冬藤提取物低剂量组、中剂量组、高剂量组。除对照组外均建模,建模成功后,低、中、高剂量组分别给予50、100、200 mg·kg^(-1)·d(-1)忍冬藤提取物,灌胃12周,对照组和模型组给予等体积生理盐水。对比各组双能X线摄片结果、骨痂组织H-E染色结果、骨密度、血清碱性磷酸酶(ALP)水平、血清炎症因子即肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)水平,应用免疫印迹和RT-PCR检测骨痂组织Wnt3、β-catenin蛋白和mRNA表达水平。结果:双能X线摄片结果显示,模型组骨折线清晰、骨痂生长明显;与模型组相比,低、中、高剂量组骨折端缝隙以及骨痂体积减小,高剂量组骨折愈合最佳。H-E染色结果显示,模型组可见软骨性骨痂,成熟的骨小梁细胞较少,骨小梁排列紊乱;低、中、高剂量组,组织病理变化有程度不同改善,骨小梁数目逐渐增多,排列趋于有序。与对照组比较,模型组大鼠骨密度、ALP及Wnt3、β-catenin蛋白和mRNA表达水平显著降低,血清TNF-α、IL-1β水平显著升高;与模型组比较,低、中、高剂量组骨密度、ALP及Wnt3、β-catenin蛋白和mRNA表达水平显著升高,血清TNF-α、IL-1β水平显著降低。结论:忍冬藤提取物可促进骨质疏松性骨折大鼠模型骨折愈合,其机制可能与其能激活Wnt/β-catenin通路有关。Objective:To investigate the effects of honeysuckle vine extract on rat models of osteoporotic fractures and its mechanism.Methods:The rats were randomly divided into Control group,Model group,and low-dose(Low),medium-dose(Medium),and high-dose(High) honeysuckle vine extract groups.Modeling was performed in all groups except for the Control group.After successful modeling,the Low,Medium,and High groups were given 50,100,and 200 mg · kg^(-1)· d^(-1) honeysuckle vine extract by gavage for 12 weeks,while the Control group and Model group were given equal volume of normal saline.The dual energy X-ray radiography results,callus H-E staining results,bone density,serum alkaline phosphatase(ALP) levels,and serum inflammatory cytokine [tumor necrosis factor-alpha(TNF-α),interleukin-1 beta(IL-1β)] levels were measured and compared between the groups.Western blotting and reverse transcription-polymerase chain reaction(RT-PCR) were used to detect the Wnt3,β-catenin protein,and mRNA expression levels in callus tissues.Results:The results of dual energy X-ray radiography showed that the fracture line of the Model group was clear,and there was evident callus growth.Compared with the Model group,the Low,Medium and High groups showed reduced gap at the fracture site and smaller callus volume.Among them,the High group exhibited the best fracture healing.H-E staining results showed that chondrogenic callus was observed in the Model group,with fewer mature trabecular cells and disordered bone trabeculae.In the Low,Medium and High groups,the histopathological changes were improved to varying degrees,with an increasing number of trabeculae and their arrangement became more orderly.Compared with the Control group,the Model group of rats showed significant decreases in bone density,ALP levels,as well as expression levels of Wnt3 and β-catenin proteins and m RNA.Additionally,serum levels of TNF-α and IL-1β were significantly elevated.Compared with the Model group,the Low,Medium,and High groups showed significant increases in bone
