基于HPLC-Q-TOF-MS技术的姜半夏及其伪品姜水半夏的鉴别研究

摘  要:目的基于高效液相色谱四极杆飞行时间质谱(high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry,HPLC-Q-TOF-MS)联用技术结合化学计量学建立姜半夏和姜水半夏的鉴定方法。方法采用Agilent Poroshell 120 SB-C_(18)(2.1×150 mm,2.7μm)色谱柱,以甲醇-5 mmol/L乙酸铵溶液为流动相,0.3 mL/min流速梯度洗脱。电喷雾离子化源、QTOF作为检测器,正离子模式检测。Profinder软件进行特征化合物提取。将Profinder软件导出的数据以PFA格式导入安捷伦代谢组学统计分析软件(mass profiler professional,MPP)。通过MPP软件的主成分分析,查找姜半夏和姜水半夏之间有显著性变化的化合物,以偏最小二乘法建立姜半夏真伪鉴别预测模型,预测样品的掺伪。结果筛选得到姜水半夏特有化合物237个,姜半夏特有化合物242个。建立了姜半夏真伪鉴别模型,在掺入一定比例姜水半夏时,模型能预测样品偏离,具有一定的预测性。结论该方法可为判别姜半夏的掺伪提供参考依据。Objective To establish a method for identification of Pinelliae Rhizoma Praeparatum cum Zingibere et Alumine(PRPZA)and Rhizoma Typhonii Flagelliformis cum Zingibere et Alumine(RTFZA)by high performance liquid chromatography quadrupole time-of-flight mass spectrometry(HPLC-Q-TOF-MS)combined with chemometrics.Methods Chromatograpic separation was performed on Agilent Poroshell 120 SB-C_(18)(2.1×150 mm,2.7μm)column with a mobile phase of methanol and 5 mmol/L ammonium acetate at a flow rate of 0.3 mL/min for gradient elution.QTOF equipped with ESI ion source was performed in positive ionization.Profinder software was used to extract characteristic compounds.The data exported from Profinder software were imported into Agilent metabolomics statistical analysis software(mass profiler professional,MPP)in PFA format.Through the principal component analysis of MPP software,the compounds with significant changes between PRPZA and RTFZA were found,and the partial least squares method was used to establish the authenticity identification prediction model of PRPZA to predict the adulteration of samples.Results A total of 242 compounds specific to PRPZA and 237 compounds specific to RTFZA were screened.The authenticity identification model of PRPZA was established.When a certain proportion of RTFZA was mixed,the model could predict the deviation of the sample and had certain predictability.Conclusion This method can provide a reference for judging the adulteration of PRPZA.
