摘  要:为研究七叶树提取物(ACE)对皮肤的修护功效,采用辣椒素(CAP)刺激稳定高表达瞬时感受器电位受体V1(TPRV1)的HaCaT-TRPV1-OE细胞,验证其对TPRV1的作用;采用脂多糖(LPS)刺激血管内皮细胞(HUVEC),验证其对内皮素(ET-1)和细胞间粘附分子(ICAM-1)表达的影响。同时将其配制成精华乳,于面部皮肤上使用28 d后,通过探头检测、医生评分评价其对眼周色素沉着、眼袋,以及面颊泛红的影响。结果显示,ACE在0.031%~0.125%浓度下,对CAP诱导的TRPV1活化具有抑制作用,可显著减少钙离子内流,也可降低LPS诱导的HUVEC细胞活化,显著减少ET-1表达量,减少ICAM-1表达。面部皮肤上,2%的ACE可减少眼下色素沉着、增加真皮层胶原密度,降低黑眼圈、眼袋评分,减少皮肤红度,改善面颊泛红症状。因此,七叶树提取物具有修护功效。In order to study the repair efficacy of Aescus chinensis extract(ACE),capsaicin(CAP)was used to stimulate HaCaT-TRPV1-OE cells with stable and high expression of transient receptor potential receptor V1(TPRV1)to verify its inhibitory effect on TPRV1.In addition,lipopolysaccharide(LPS)-stimulated vascular endothelial cell(HUVEC)model was used to verify its effect on the expression of endothelin(ET-1)and intercellular adhesion molecule(ICAM-1),which are the key factors in capillaries.Meanwhile,after clinically applied for 28 days,its effects on under-eye pigmentation and eye bags,as well as cheek redness were evaluated by probe detection and dermatologist scoring.The results showed that at the concentration of 0.031%~0.125%,ACE inhibited the activation of TRPV1 induced by CAP by significantly reducing the influx of calcium,and decreased the activation of HUVEC cells induced by LPS by significantly reducing the expression of ET-1 and ICAM-1.On facial skin,2%concentration of ACE could reduce the under-eye pigmentation,increase the dermal collagen density,reduce the score of dark circles and eye bags,reduce skin a*value,and improve the symptoms of cheek redness.Therefore,ACE has skin repair efficacy.
