摘 要:目的探讨采用“肺炎支原体培养液滴鼻+烟熏法+辣椒素溶液雾化”建立肺炎支原体肺炎(mycoplasma pneumoniae pneumonia,MPP)感染后痉咳大鼠模型的可能性。方法40只SPF级雄性Wistar大鼠随机分为空白组、MPP组、烟熏组、MPP痉咳组,采用“肺炎支原体培养液滴鼻+烟熏法+辣椒素溶液雾化”建立MPP感染后痉咳大鼠模型,观测并记录各组大鼠一般行为学表现、咳嗽次数、大鼠支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中白细胞总数,观察肺组织病理学改变情况。结果实验过程中,空白组大鼠行为表现正常。随着烟熏天数的增加,各模型组大鼠表现为精神萎靡不振,呼吸声嘶哑,个别大鼠出现间断性咳嗽等经典表现。辣椒素溶液雾化吸入激发后,各模型组大鼠出现明显咳嗽症状,表现为颈部向前、口张开、耳朵战栗等。与空白组比较,MPP痉咳组大鼠咳嗽次数明显增多,差异有统计学意义(P<0.05);MPP组、烟熏组大鼠咳嗽次数有所增加,但差异无统计学意义(P>0.05)。与空白组比较,MPP组、烟熏组、MPP痉咳组大鼠BALF中白细胞总数均显著升高,差异有统计学意义(P<0.05);3个模型组两两比较,差异无统计学意义(P>0.05)。结论采用“肺炎支原体培养液滴鼻+烟熏法+辣椒素溶液雾化”建立MPP感染后痉咳大鼠模型的方法切实可行。Objective e To investigate the possibility of establishing a rat model of spasmodic cough after myco-plasma pneumoniae pneumoniae(MPP)infection by using"mycoplasma pneumoniae culture solution intranasal in-stillation+smoking exposure+capsaicin solution nebulisation".Methods 40 SPF-grade male Wistar rats were ran-domly divided into four groups:control group,MPP group,smoking exposure group,and MPP spasmodic cough group.The MPP spasmodic cough rat model was established by using the combined approach of"mycoplasma pneu-moniae culture solution intranasal instillation+smoking exposure+capsaicin solution nebulization".General behavior-al manifestations,cough frequency,total white blood cell count in bronchoalveolar lavage fluid(BALF),and patho-logical changes in lung tissue were observed and recorded in each group.Results During the experiment,the con-trol group rats exhibited normal behavioral manifestations.With increasing days of smoking exposure,rats in the mod-el groups showed symptoms such as lethargy,hoarse breathing,and intermittent cough in some individuals.After capsa-icin solution nebulization,rats in all model groups exhibited obvious cough symptoms,characterized by neck exten-sion,mouth opening,and ear tremor.Compared with the control group,the MPP spasmodic cough group had a signifi-cantly increased cough frequency,with a statistically significant difference(P<0.05).The MPP group and smoking ex-posure group showed an increased cough frequency,but the difference was not statistically significant(P>0.05).Com-pared with the control group,the total white blood cell count in BALF was significantly increased in the MPP group,smoking exposure group,and MPP spasmodic cough group,with a statistically significant difference(P<0.05).Howev-er,there was no significant difference between the three model groups(P>0.05).Conclusion It is feasible to estab-lish a rat model of spasmodic cough after mycoplasma pneumoniae pneumoniae(MPP)infection by using"mycoplas-ma pneumoniae culture solution intranasal instllation+smoking
