摘 要:研究旨在通过体外培养试验探究在饲粮中添加黑沙蒿醇提物对奶牛瘤胃内环境的影响,以筛选黑沙蒿醇提物的最适添加剂量。试验选用4头健康、泌乳期相同、体况相近的荷斯坦奶牛采集口腔瘤胃液,作为瘤胃液的供体。体外培养试验分为4组,对照组以精粗比4∶6的饲粮为发酵底物,不添加黑沙蒿醇提物,试验组在发酵底物中分别添加0.03%(试验1组)、0.3%(试验2组)和3.0%(试验3组)的黑沙蒿醇提物(风干基础)。分别培养至0、3、6、9、12、24 h,记录产气量并采集培养液样品,测定pH、氨态氮(NH_(3)-N)浓度和挥发性脂肪酸(VFA)含量等瘤胃发酵参数,并评定多项指标综合指数(MFAEI)。结果表明:随着培养时间的延长,各组产气量均上升。除试验1组和试验2组在培养至3~6 h时,pH先升高后降低,其余各组瘤胃液pH均呈下降趋势,且在正常范围内(6.0~7.5)。培养至3~12 h时,各试验组NH_(3)-N浓度均显著高于对照组(P<0.05);培养至24 h,试验2组和试验3组NH_(3)-N浓度均低于对照组。各试验组挥发性脂肪酸(VFA)浓度较对照组有所降低,而试验2组与对照组变化趋势一致,二者均优于试验1、3组。说明在黑沙蒿醇提物添加剂量为0.3%时对促进奶牛体外瘤胃发酵功能的效果较好。The aim of this study was to investigate the effects of Artemisa ordosica alcohol extract on rumen environment of dairy cows by in vitro culture experiment,and to screen the optimal dosage of Artemisa ordosica alcohol extract.The rumen fluid was collected from four healthy Holstein cows with the same lactation period and similar body condition as the rumen fluid donor.In vitro culture experiment was divided into 4 groups.The control group was fed with a 4∶6 ratio of concentrate to crude as the fermentation substrate without Artemisa ordosica alcohol extract,and the experimental groups were added 0.03%(experiment group 1),0.3%(experiment group 2)and 3.0%(experiment group 3)Artemisa ordosica alcohol extract,respectively(DM basis).Gas production was respectively recorded after fermented for 0,3,6,9,12 and 24 h,and the culture solution samples were collected.Rumen fermentation parameters such as pH,NH_(3)-N concentration and VFA content were determined,and evaluate the comprehensive index of multiple index(MFAEI).The results showed as follows:with the extension of culture time,the gas production of each group increased.Except for experiment group 1 and experiment group 2,the pH of rumen fluid in other groups showed a downward trend and was in the normal range(6.0-7.5)after 3-6 h of culture.The concentration of NH_(3)-N in each experimental group was significantly higher than that in the control group from 3-12 h,and the concentration of NH_(3)-N in the experiment group 2 and experiment group 3 were lower than that in the control group at 24 h.Each experiment group significantly reduced the concentration of volatile fatty acids(VFA)in each component,and the changes in the experiment group 2 were comparable to the control group and both were better than the other two dose groups.According to the change of rumen parameters,combined with the multiply factor associative effects index result,it is more suitable to promote the rumen fermentation function of dairy cows in vitro when the dosage of Artemisa ordosica alcohol
