摘 要:利用肿瘤偏酸性微环境,构建黄芩苷pH敏感脂质体。以包封率为评价指标,采用Plackett-Burman实验设计筛选处方工艺,并采用Box-Behnke响应面实验确定最佳处方工艺,进而对制剂进行表征,考察其在不同pH介质中的体外释放特性,筛选出黄芩苷长循环脂质体的最佳制备工艺,并对制剂进行表征,考察其体外释放度行为。结果表明最优工艺及处方制备出的脂质体,平均粒径97.56 nm,包封率84.92%,其体外释放具有缓释性,且具有pH敏感性。Utilizing the tumor's acidic microenvironment,pH-sensitive liposomes of baicalin were constructed.The encapsulation efficiency was used as a criterion for evaluation,and Plackett-Burman experimental design was employed to screen the formulation process.Subsequently,Box-Behnken response surface methodology was used to determine the optimal formulation process.The resulting formulation was characterized,and its in vitro release characteristics in different pH media were investigated to identify the optimal preparation process for long-circulating liposomes of baicalin,followed by characterization and examination of its release behavior.The results indicated that the liposomes prepared using the optimal process and formulation had an average particle size of 97.56 nm and an encapsulation efficiency of 84.92%.The in vitro release exhibited sustained-release properties and pH sensitivity.
