MS-222对墨瑞鳕麻醉效果及血清生化指标的影响

摘　　要：为探索MS-222麻醉剂对墨瑞鳕(734.94±51.36 g)的麻醉效果,在水温18±0.5℃条件下,采用不同浓度(0.10、0.15、0.20、0.30 mg·L^(-1))麻醉剂进行麻醉实验,并分析麻醉复苏后血清生化指标的变化。结果显示,在麻醉及复苏时间效果上,麻醉有效浓度范围为0.185~0.227 mg·L^(-1)。麻醉期间,血清中的葡萄糖、皮质醇含量与对照组相比均存在显著差异(P<0.05),随麻醉剂浓度增加,葡萄糖含量先升高后降低,皮质醇含量先降低再升高;碱性磷酸酶活性和谷丙转氨酶活性仅在0.30 mg·L^(-1)组显著升高(P<0.05);超氧歧化酶活性先降低后升高,过氧化氢酶活性逐渐下降。复苏12 h后,葡萄糖含量、谷丙转氨酶活性、超氧歧化酶活性均恢复到对照组水平,而皮质醇含量除0.10 mg·L^(-1)组、碱性磷酸酶活性及过氧化氢酶活性除0.30 mg·L^(-1)组外,其他组均恢复到对照组水平。综合考虑MS-222麻醉剂的麻醉效果及对墨瑞鳕机体的影响,麻醉剂浓度在0.20 mg·L^(-1)具有最佳效果。研究结果可为MS-222麻醉剂使用安全性及其麻醉机理的分析提供参考依据。Maccullochella peelii with mean body weight of 734.94±51.36 g was exposed to MS-222 at a dose of 0.10,0.15,0.20,0.30 mg·L^(-1)in order to explore the anaesthetic effect and the changes of serum biochemical indexes after anesthesia and resuscitation,at the water temperature of 18±0.5℃.The results showed that the effective concentration range of MS-222 was 0.185-0.227 mg·L^(-1)considering the time required for anesthesia and resuscitation.During anesthesia,contents of glucose(GLU)and cortisol(COR)in serum were significantly different compared with control group(P<0.05).With the increase of anesthetic concentration,the glucose content first increased and then decreased,while the change of cortisol was the opposite;The activity of Alkaline phosphatase(ALP)and Alanine aminotransferase(ALT)were significantly increased in 0.30 mg·L^(-1)group(P<0.05);The activity of Superoxide dismutase(SOD)decreased first and then increased,while catalase(CAT)decreased gradually.After 12 hours of resuscitation,except for the 0.10 mg·L^(-1)group of COR and the 0.30 mg·L^(-1)group of ALP and CAT,all other groups recovered to the control group level.Considering the anesthetic effect of MS-222 and its influence on M.peelii,the anesthetic concentration of 0.20 mg·L^(-1)in this experiment was the best.The results of this study can provide reference for the analysis of safety of MS-222 and its anesthesia mechanism.
