摘 要:目的:观察藏红花素对桥本甲状腺炎(HT)模型大鼠甲状腺自身抗体及甲状腺激素水平的影响。方法:将21只SD大鼠随机分为3组,每组7只。空白对照组予蒸馏水常规喂养,造模组制备HT模型,同时予等量蒸馏水灌胃处理。藏红花素组制备HT模型同时予藏红花素20 mg/kg灌胃处理,每天1次。第8周采用ELISA试剂盒检测血清中抗甲状腺过氧化物酶抗体(TPO-Ab)、抗甲状腺球蛋白抗体(TG-Ab)、游离三碘甲腺原氨酸(FT3)、游离甲状腺素(FT4)及氧化应激标志物活性氧簇(ROS)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)水平。结果:与空白对照组比较,造模组血清中TPO-Ab、TG-Ab、FT3、FT4、ROS含量上升(P<0.05),SOD、GSH含量下降(P<0.05)。与造模组比较,藏红花素组血清中TPO-Ab、TG-Ab、FT3、FT4、ROS含量下降(P<0.05),SOD、GSH含量上升(P<0.05)。经Pearson相关分析结果显示,血清SOD、GSH与TPO-Ab、TG-Ab水平均呈负相关(P<0.001)。ROS和TPO-Ab、TG-Ab之间呈正相关(P<0.001)。结论:藏红花素可以有效降低HT大鼠氧化应激水平,抑制大鼠TPO-Ab、TG-Ab的升高,改善甲状腺功能紊乱状态,其作用机制可能与其抗氧化应激相关。Objective:To observe the effects of crocin on thyroid autoantibodies and thyroid hormone levels in Hashimoto thyroiditis(HT)model rats.Methods:A total of 21 SD rats were randomly divided into three groups,with seven rats in each group.The blank control group was fed with distilled water;the model group was prepared with HT model,and given intragastric treatment of the same amount of distilled water.The crocin group was prepared with HT model,and was given intragastric treatment of 20 mg/kg of crocin once a day.At the 8th week,serum levels of anti-thyroid peroxidase antibody(TPO-Ab),antithyroglobulin antibody(TG-Ab),free triiodothyronine(FT3),free thyroxine(FT4),oxidative stress markers reactive oxygen species(ROS),superoxide dismutase(SOD),glutathione(GS)were detected by ELISA.Results:Compared with those in the blank control group,the levels of serum TPO-Ab,TG-Ab,FT3, FT4 and ROS in the model group were increased (P<0.05), while the levels of SOD and GSH weredecreased (P<0.05). Compared with those in the model group, the levels of serum TPO-Ab, TG-Ab,FT3, FT4 and ROS in the crocin group were reduced (P<0.05), while the levels of SOD and GSH wereenhanced (P<0.05). The results of Pearson correlation analysis showed that the levels of serum SOD andGSH were negatively correlated with TPO-Ab and TG-Ab (P<0.001). and that ROS was positivelycorrelated with TPO-Ab and TG-Ab (P<0.001). Conclusion: Crocin can effectively reduce the oxidativestress levels in HT rats,inhibit the increase of TPO-Ab and TG-Ab,and improve the thyroid dysfunction.The mechanism of action may be related to the anti-oxidative stress.
