摘　　要：目的探讨含PE ST的核蛋白(PEST-containing nuclear protein,PCNP)在甲状腺癌(thyroid carcinoma,TC)中的表达,并分析其表达BRAF V600E基因突变的相关性及临床意义。方法采用免疫组化法检测110例TC与其癌旁组织及38例甲状腺肿组织中PCNP的表达水平,并利用荧光定量PCR检测其中65例TC患者中BRAF V600E基因突变情况,探究两者的关系及临床意义。结果单因素分析显示在110例TC中PCNP的表达与患者的性别、年龄、淋巴结有无转移、组织学分型相关,P<0.05。其中57例甲状腺乳头状癌中PCNP的表达明显高于其癌旁组织及甲状腺肿组织的表达,P<0.001。Pearson相关分析显示65例TC中PCNP的表达与BRAF V600E基因突变无相关性。结论PCNP在TC的表达有重要临床意义;PCNP与BRAF V600E基因突变在甲状腺中的相关性分析证明二者在TC发生、发展过程中可能分别起到不同的作用。Objective To investigate the expression of PEST-containing nuclear protein(PCNP)in thyroid cancer and analyze its correlation and clinical significance of BRAF V600E gene mutation.Methods Immunohistochemistry was used to detect PCNP expression in 110 cases of TC,its adjacent tissues and 38 cases of goitre tissues,and fluorescence quantitative PCR was used to detect BRAF V600E gene mutation in 65 cases of TC patients to explore the relationship and clinical significance of the two.Results Univariate analysis showed that the expression of PCNP in 110 TC patients was correlated with gender,age,lymph node metastasis and tissue type,and the differences were statistically significant(P<0.05).The expression of PCNP in 57 cases of papillary thyroid carcinoma was significantly higher than that in adjacent tissues and goiter tissues,with statistical significance(P<0.001).Pearson correlation analysis showed that there was no correlation between PCNP expression and BRAF V600E gene mutation in 65 TC patients(P>0.05).Conclusion The expression of PCNP in TC has important clinical significance.The correlation analysis of PCNP and BRAF V600E gene mutations in thyroid gland indicated that they may play different roles in the occurrence and development of TC.
