摘 要:目的:探讨盆腔炎Ⅱ号方治疗盆腔炎性疾病后遗症(Sequela of Pelvic Inflammatory Disease, SPID)的作用和机制。方法:将30只SPID大鼠模型随机分为假手术组、模型组、盆腔炎Ⅱ号方低剂量组、盆腔炎Ⅱ号方中剂量组、盆腔炎Ⅱ号方高剂量组,每组6只。运用酶联免疫吸附法检测大鼠子宫组织中炎症细胞因子的表达水平,Masson染色观察组织病理学改变,免疫组化法检测Ⅰ型胶原蛋白和Ⅲ型胶原蛋白表达水平,蛋白免疫印迹法检测转化生长因子-β1(Transforming Growth Factor-β1,TGF-β1)/Smad2信号通路相关蛋白表达。结果:与模型组相比,盆腔炎Ⅱ号方干预可显著降低大鼠子宫组织中白细胞介素(Interleukin,IL)-1β、IL-6和肿瘤坏死因子(Tumor Necrosis Factor,TNF)-α表达水平,减少胶原蛋白沉积及Ⅰ型胶原蛋白、Ⅲ型胶原蛋白表达,下调TGF-β1和p-Smad2蛋白表达(P <0.05)。结论:盆腔炎Ⅱ号方能缓解SPID大鼠子宫组织炎症反应,抑制TGF-β1/Smad2信号通路,改善组织纤维化。Objective:To explore the effects and mechanisms of pelvic inflammation formulation No.2 on sequela of pelvic Oinflammatory disease(SPID).Methods:Atotal of 30 rat models of SPID were randomly divided into the sham group,model group,low-dose,middle-dose,and high-dose pelvic inflammation formulation No.2 groups with 6 rats in each group.Enzyme-linked immunosorbent assay was utilized to examine the levels of inflammatory cytokines in the uterine tissues of SPID rats.The histopathological changes were observed using Masson staining.The expressions of Collagen 1 and Collagen 3 were detected by immunohistochemical staining.The transforming growth factor-β1(TGF-β1)/Smad2 signaling pathway was evaluated by Western blot analysis.Results:Comparing to the model group,pelvic inflammation formulation No.2 treatment dramatically decreased the levels of interleukin-1β(IL-1β),IL-6 and tumor necrosis factor-α,inhibited the deposition of collagen proteins and the expressions of Collagen 1 and Collagen 3,downregulated the protein levels of TGF-β1 and p-Smad2(P<0.05).Conclusion:Pelvic inflammation formulation No.2 was capable of ameliorating the inflammatory response in the uterine tissues of SPID rats and restricting fibrogenesis by suppression of the TGF-β1/Smad2 signaling pathway.
