摘  要:目的建立同时测定肉桂不同部位挥发油中肉桂酸、桂皮醛、邻甲氧基肉桂醛含量的HPLC-RID方法,并比较它们的体外透皮特性。方法采用Franz扩散池法,以小鼠皮肤作渗透介质,分别对桂皮、桂枝、桂叶的挥发油进行体外透皮吸收试验,采用Agilent C18色谱柱(4.6 mm×250 mm,5μm),乙腈-0.2%磷酸(30∶70)为流动相,流速0.9 mL·min^(-1),示差折光检测器(RID)温度30℃,柱温30℃,同时测定透皮样品中肉桂酸、桂皮醛、邻甲氧基肉桂醛的含量,计算并分析各成分的累计渗透量和渗透速率。结果肉桂酸、桂皮醛、邻甲氧基肉桂醛分别在0.0489~0.5381μg、0.1816~9.9874μg、0.1142~1.2566μg与峰面积线性关系良好(r>0.999);三种成分的24 h单位面积累计渗透量,桂皮油的为53.84、1228.10、265.29μg·cm^(-2),桂枝油的为84.73、2937.69、87.39μg·cm^(-2),桂叶油的为67.45、1699.78、347.99μg·cm^(-2);三种成分的稳态渗透速率,桂皮油的为1.41、96.24、12.77μg·cm^(-2)·h^(-1),桂枝油的为1.97、130.01、4.90μg·cm^(-2)·h^(-1);桂叶油的为0.91、74.85、16.48μg·cm^(-2)·h^(-1)。结论建立的HPLC-RID方法快速简便,分离度好,准确度高,可用于肉桂油的质量控制及其透皮试验研究。挥发油中各指标成分均能透过小鼠离体皮肤,24 h累计渗透量和渗透速率的大小顺序均是桂皮醛>邻甲氧基肉桂醛>肉桂酸,且与成分的浓度成正相关。Objective To establish a method to simultaneously determine the content of cinnamic acid,cinnamaldehyde and 2-methoxy cinnamaldehyde by HPLC-RID,and to compare the in vitro transdermal properties of volatile oil from different parts of Cinnamomum cassia.Methods Mouse skin was used as the permeation medium,the in vitro transdermal absorption experiments of cinnamon bark oil,cassia twig oil and cinnamon leaf oil were conducted with Franz diffusion cells.We simultaneously determinated the content of cinnamic acid,cinnamaldehyde and 2-methoxy cinnamaldehyde in transdermal samples by Agilent C18 column(4.6 mm×250 mm,5μm).Acetonitrile-0.2%phosphoric acid(30∶70)was used as the mobile phase,and the flow rate were all at 0.9 mL·min^(-1).The refractive index detector(RID)was used,and the temperatures of column and detector were all at 30℃.The accumulative permeation and rate of each component were calculated and analyzed.Results Cinnamic acid,cinnamaldehyde and 2-methoxy cinnamaldehyde were linear within 0.0489~0.5381μg,0.1816~9.9874μg,and 0.1142~1.2566μg,respectively(r>0.999).The 24 h accumulative permeation per unit area was 53.84,1228.10,and 265.29μg·cm^(-2)from the cinnamon bark oil;84.73,2937.69,and 87.39μg·cm^(-2)from the cassia twig oil;while 67.45,1699.78,and 347.99μg·cm^(-2)from the cinnamon leaf oil.The steady-state permeation rates were 1.41,96.24,and 12.77μg·cm^(-2)·h^(-1)from the cinnamon bark oil;1.97,130.01,and 4.90μg·cm^(-2)·h^(-1)from the cassia twig oil;and 0.91,74.85,and 16.48μg·cm^(-2)·h^(-1)from the cinnamon leaf oil.Conclusion The established HPLC-RID method is fast,simple,with good separation and high accuracy,which can be used for the quality control and transdermal testing of cinnamon oil.All these indicator components in these volatile oils can permeate the mice skin in vitro.The accumulative permeation and rate from largest to smallest are cinnamaldehyde,2-methoxy cinnamaldehyde and cinnamic acid.
