摘 要:目的:运用响应面Box-Behnken实验设计(Box-Behnken Design,BBD)法分析提取时间、功率、温度和液料比4种因素对红瑞木果多糖提取率的影响,优化超声辅助提取工艺。方法:首先进行超声辅助提取红瑞木果多糖的单因素试验,获得提取时间、功率、温度和液料比的最佳范围,进一步利用BBD建造模型,进行多糖提取率的测定,建立回归模型,进行方差分析,然后构建响应面和等高线图,最后进行最优参数的检验,从而确定4种因素的影响大小和最佳提取条件。结果:单因素实验显示,当温度、时间、功率和液料比分别为50℃、15 min、400 W和(20∶1)mL/g时,多糖提取率最大;拟合分析方程显示,影响红瑞木果多糖提取率的因素依次为提取功率>时间>温度>液料比;选取提取温度(40℃、50℃、60℃)、时间(10 min、15 min、20 min)、功率(300 W、400 W、500 W)、液料比(15∶1、20∶1、25∶1) mL/g进行响应面与等高线图分析,每两个因素的交互作用对红瑞木果多糖提取率的影响均呈椭圆形,交互作用均明显;最佳提取工艺条件显示,当温度、时间、功率和液料比分别为50℃、17 min、416 W、(19∶1)mL/g时,提取率为6.36%,与BBD模型的预测结果相当。结论:采用响应面BBD法所优化的红瑞木果多糖超声提取工艺具有良好的预测效果和较高的提取率,值得推广应用。Objective:Using response surface methodology(BBD)to analyze the effects of extraction time,power,temperature,Oand liquid to material ratio on the extraction rate of polysaccharides from swida alba opiz,and optimizing the ultrasound assisted extraction process.Methods:Firstly,a single factor experiment on ultrasound assisted extraction of polysaccharides from swida alba opiz was conducted to obtain the optimal ranges of extraction time,power,temperature,and liquid to material ratio.BBD was used to construct a model to measure the polysaccharide extraction rate,a regression model was established to perform variance analysis.Then response surfaces and contour maps were conducted.Finally,the optimal parameters were tested to determine the impact of the four factors and the optimal extraction conditions.Results:The single factor experiment showed that the polysaccharide extraction rate was highest when the temperature,time,power,and liquid to material ratio were 50℃,15 min,400 W,and(20:1)mL/g,respectively.The fitting analysis equation showed that the factors that affected the extraction rate of polysaccharides from swida alba opiz were in the following order:extraction power>time>temperature>liquid to material ratio;selecting extraction temperature(40℃,50℃,60℃),time(10 min,15 min,20 min),power(300 W,400 W,500 W),and liquid to material ratio(15:1,20:1,25:1)mL/g for response surface and contour plot analysis,the interaction between each two factors had an elliptical effect on the extraction rate of polysaccharides from swida alba opiz,and the interaction was significant;the optimal extraction process conditions showed that the extraction rate was 6.36%when the temperature,time,power,and liquid to material ratio were 50℃,17 min,416 W,and(19:1)mL/g,respectively,which was consistent with the predicted results of the BBD model.Conclusion:The optimized ultrasonic extraction process of polysaccharides from swida alba opiz using response surface BBD method has good predictive effects and high extraction rate,which is
