柔嫩艾美耳球虫棒状体颈部蛋白EtRON2_(L2)特性与功能初步分析

摘　　要：利用RT-PCR、间接免疫荧光实验、免疫印迹实验、抗体中和实验以及BiFC等方法对柔嫩艾美耳球虫棒状体颈部蛋白2同源分子—Et RON2_(L2)的特性和功能进行研究。结果发现Et RON2_(L2)氨基酸序列与常规的Et RON2仅有26.63%的同源性,mRNA水平在第二代裂殖子中最高,而蛋白水平在第二代裂殖子和子孢子阶段最高;Et RON2_(L2)在游离的第二代裂殖子和子孢子阶段主要分布于胞质,而在入侵后子孢子中主要分布于虫体表面;抗rEt RON2_(L2)多克隆血清处理组明显抑制子孢子入侵细胞;Et RON2_(L2)与EtAMA3之间存在互作关系。本研究为探究Et RON2_(L2)在球虫入侵过程中的作用机制奠定基础。The molecular characteristics and biological function of rhoptry neck protein RON2L2 of Eimeria tenella(EtRON2_(L2))were investigated in the present study using RT-PCR,indirect immunofluorescence assay,Western blot,antibody neutralization and BiFC.The results showed that the amino acid sequence of EtRON2_(L2)had 26.63%identity with Et RON2.The mRNA level of EtRON2_(L2)was the highest in the second generation merozoites while the translation level was higher in both the second generation merozoites and sporozoites.EtRON2_(L2)was mainly distributed in the cytoplasm of the free second generation merozoites and sporozoites while it was mainly present on the surface of the parasites after invading cells.The anti-rEtRON2_(L2)antibodies had a significant inhibitory effect on the invasion of sporozoites.At the same time,there was an interaction between EtRON2L2 and E.tenella apical membrane antigen 3(EtAMA3).The results of this study laid a foundation for further research on the function and mechanism of EtRON2_(L2)in the invading process of E.tenella.
