摘 要:胀果甘草是种植面积最广且适宜新疆气候的一种甘草品种。盐胁迫条件会特异性诱导胀果甘草中部分基因的表达,在转录组水平上研究差异表达基因应对胁迫的表达模式,更有利于阐明抗逆调控的分子机制。本研究利用高通量测序技术Illumina2000对盐胁迫处理后的胀果甘草根系进行转录组测序,鉴定到2866个诱导上调表达的基因,1321个诱导下调表达的基因。对所有差异表达基因进行GO分析发现,这些基因主要参与酶活性变化、离子结合、氧化还原过程、防御反应等。此外,通过KEGG分析发现,差异表达基因主要富集在核糖体、氧化磷酸化、疾病调控等代谢通路。本研究对胀果甘草抗盐基因挖掘和今后采用分子生物学手段培育甘草新品种具有重要意义。Glycyrrhiza inflata is a kind of glycyrrhiza with the widest planting area and suitable for Xinjiang climate.Salt stress can specifically induce the expression of some genes in Glycyrrhiza inflate.Studying the expression patterns of differentially expressed genes in response to stress at the transcriptome level is more conducive to elucidate the molecular mechanism of stress resistance regulation.In this study,2866 up-regulated genes and 1321 down-regulated genes were identified by high-throughput sequencing technology Illumina2000 to sequence the transcriptome of licorice(glycyrrhiza uralensis)roots after salt stress treatment.Gene Ontology(GO)analysis of the differentially expressed genes revealed their predominant involvement in various biological processes,including enzyme activity alteration,ion binding,REDOX process,defense response,etc.KEGG enrichment analysis showed that differentially expressed genes were mainly concentrated in ribosome,oxidative phosphorylation and disease regulation pathways.This study is of great significance for the discovery of salt-resistant genes and the cultivation of new varieties of Glycyrrhiza inflata by molecular biological methods in the future.
