摘 要:目的研究迷迭香酸(RosA)体外抗水痘-带状疱疹病毒(VZV)的作用及其机制。方法采用不同浓度的RosA(400、200、100、50μmol/L)处理VZV感染的Vero细胞,采用细胞病变效应(CPE)法和MTT法评价RosA体外抗VZV的作用以及对细胞的毒性,采用RT-qPCR法检测RosA对VZV ORF36 mRNA表达的影响。结果当RosA浓度为100、200、400μmol/L时,RosA对VZV的抑制率分别为55.88%、78.99%和98.19%,其抑制效果与RosA浓度呈正比。当RosA浓度为400μmol/L时,RosA对VZV的灭活率为85.44%,灭活作用的EC_(50)为185.95μmol/L,RosA的CC 50为2362.10μmol/L,EC_(50)为63.02μmol/L,SI为37.48。当RosA浓度为100、200、400μmol/L时,VZV ORF36 mRNA的表达量分别下调了48.2%、85.9%、97.2%。结论RosA在体外具有抗VZV的作用,其抗病毒作用与RosA抑制VZV ORF36的转录有关。Objective To study the anti-varicella-zoster virus(VZV)effect of rosmarinic acid(RosA)in vitro and its mechanism.Methods VZV-infected Vero cells were treated with different concentrations of RosA(400,200,100,and 50μmol/L).The anti-VZV effect and cytotoxicity of RosA in vitro were evaluated by cytopathic effect(CPE)and MTT staining,respectively.The effect of RosA on the expression of VZV ORF36 mRNA was detected by RT-qPCR.Results When the concentration of RosA was 100,200,and 400μmol/L,the inhibition rates of RosA on VZV were 55.88%,78.99%and 98.19%,respectively,which were in a concentration dependent manner.When the concentration of RosA was 400μmol/L,the inactivation rate of RosA for VZV was 85.44%.The median effective concentration(EC_(50))of the inactivation was 185.95μmol/L.The 50%cytotoxic concentration(CC 50),EC_(50)nd selection index(SI)of RosA were 2362.10μmol/L,63.02μmol/L and 37.48,respectively.When the concentration of RosA was 100,200,and 400μmol/L,the expression of VZV ORF36 mRNA was down-regulated by 48.2%,85.9%and 97.2%,respectively.Conclusion RosA has anti-VZV effect in vitro,which is related to its inhibition of VZV ORF36 transcription.
