扶正养血膏对化疗致骨髓抑制小鼠造血调控功能的影响

摘　　要：目的:观察扶正养血膏对化疗致骨髓抑制小鼠造血调控功能的影响,并通过代谢组学分析探讨其作用机制。方法:制备化疗致骨髓抑制模型小鼠,试验组灌胃给予扶正养血膏,模型组和对照组灌胃给予饮用水,比较各组小鼠外周血常规、胸腺/脾脏指数、骨髓/胸腺/脾脏病理形态等指标,采用LC-MS分析各组小鼠血清代谢谱的变化,初步探讨扶正养血膏作用机制。结果:扶正养血膏可提升化疗致骨髓抑制模型小鼠的外周血指标,对红系细胞疗效显著;可缓解化疗致骨髓抑制模型小鼠脾脏的肿大,提升小鼠骨髓造血功能;代谢组学结果显示,试验组出现5种化合物升高和44种化合物下降,可改善小鼠化疗致代谢失调。结论:扶正养血膏可改善化疗致骨髓抑制小鼠造血调控功能,缓解化疗导致的毒副反应。Objective:To observe the effect of Fuzheng yangxue ointment on hematopoietic regulatory function in chemotherapy induced myelosuppressive mice,and explore its mechanism of action through metabolomics analysis.Method:A chemotherapy induced myelosuppressive model mouse was prepared.The experimental group was given Fuzheng yangxue ointment by gavage,while the model group and control group were given drinking water by gavage.The peripheral blood routine,thymus/spleen index,bone marrow/thymus/spleen pathological morphology and other indicators of each group of mice were compared.LC-MS was used to analyze the changes in serum metabolism profiles of each group of mice,and the mechanism of action of Fuzheng yangxue ointment was preliminarily explored.Result:Fuzheng yangxue ointment can enhance the peripheral blood indicators of chemotherapy induced myelosuppressive mice,and has a significant therapeutic effect on red blood cells.It can alleviate the splenomegaly of chemotherapy induced myelosuppressive mice.and improve their bone marrow hematopoietic function.The metabolomics results showed that the experimental group showed an increase of 5compounds and a decrease of 44compounds,which can improve the metabolic disorders caused by chemotherapy in mice.Conclusion:Fuzheng yangxue ointment can improve the hematopoietic regulatory function of chemotherapy induced myelosuppressive mice and alleviate the toxic side effects caused by chemotherapy.
