清瘟败毒饮通过TLR4/MyD88/NF-κB信号通路抑制内毒素引起的小鼠急性肺损伤

摘  要:目的:研究清瘟败毒饮对内毒素引起的急性肺损伤(acute lung injury,ALI)小鼠的保护作用。方法:72只昆明小鼠,随机分为正常组、模型组、地塞米松组、清瘟败毒饮高、中、低剂量组。膜腔注射脂多糖(LPS)建立ALI模型。给药第3 d、第7 d,每组各取6只小鼠处死。采集小鼠的血液,分离小鼠肺组织,测定小鼠肺组织湿干比(湿重/干重,W/D);ELISA与PCR检测小鼠血清及肺组织的相关因子表达;观察小鼠肺组织病理变化。结果:与模型组相比,清瘟败毒饮各剂量组小鼠肺组织病理损伤减轻,肺组织W/D下降,小鼠血清的白介素1(IL-1)、肿瘤坏死因子α(TNF-α),肺组织Toll样受体4(TLR4)、髓样分化因子88(MyD88)、核因子κB(NF-κB)、TLR4 mRNA、MyD88 mRNA的表达水平降低,且不同剂量组之间呈剂量依赖性(P<0.05)。结论:清瘟败毒饮对LPS造成的小鼠急性肺损伤有保护作用。Objective:To investigate the protective effects of Qingwen Baidu Yin(QBY) on acute lung injury(ALI) induced by endotoxin in mice.Methods:A total of 72 kunming mice were randomly divided into normal group,model group,dexamethasone group,and high,medium,and low dose groups of QBY.An ALI model was established by intraperitoneal injection of lipopolysaccharide.On the 3rd and 7th days of treatment,6 mice from each group were sacrificed.Blood samples were collected,and lung tissues were isolated from the mice.The wet-to-dry ratio of lung tissues was measured.ELISA and PCR were used to detect the expression of relevant factors in serum and lung tissues;and pathological changes in the lung tissues of the mice were observed.Results:Compared with the model group,the pathological damage of lung tissues in the various dose groups of QBY was reduced,with a decrease in the wet-to-dry ratio(W/D) of lung tissues;The expression levels of interleukin-1(IL-1),tumor necrosis factor-alpha(TNF-α) in serum,and Toll-like receptor 4(TLR4),myeloid differentiation factor 88(MyD88),nuclear factor-kappa B(NF-κB),TLR4mRNA,and MyD88mRNA in lung tissues were decreased in the various dose groups,showing a dose-dependent effect among different dose groups(P<0.05).Conclusion:QBY has a protective effect on acute lung injury induced by LPS in mice.
