基于补肾益气活血法探讨加味左归丸对骨质疏松模型小鼠骨微结构的影响

摘  要:目的:探讨加味左归丸对切除双侧卵巢骨质疏松模型小鼠骨微结构的影响及其可能的作用机制,为补肾益气活血法治疗骨质疏松提供依据。方法:将21只8周龄C57BL/6J雌性小鼠随机分成模型组、假手术组、加味组,每组各7只。模型组和加味组小鼠通过切除双侧卵巢建立骨质疏松模型,假手术组小鼠切除等量脂肪组织。加味组灌胃加味左归丸溶液,模型组和假手术组灌胃等量蒸馏水。给药8周后,采用小鼠Micro-CT结合HE染色的组织切片技术,收集骨组织图像并对小梁骨进行定量分析,评估加味左归丸对骨微观结构的影响。结果:小鼠股骨骨微结构观察结果:三组小鼠骨体积分数(BV/TV,%)、骨小梁厚度(Tb.Th,mm)、骨小梁分离度(Tb.Sp,mm)总体比较,差异均有统计学意义(P<0.05);假手术组与模型组比较,骨体积分数、骨小梁厚度、骨小梁分离度差异均有统计学意义(P<0.05);加味组与模型组比较,骨体积分数、骨小梁厚度差异均有统计学意义(P<0.05),骨小梁分离度无统计学意义(P>0.05);加味组与假手术组比较,骨体积分数、骨小梁厚度、骨小梁分离度差异均无统计学意义(P>0.05)。小鼠股骨组织形态观察结果:三组小鼠骨小梁面积百分比(%)总体比较,差异有统计学意义(P<0.05),假手术组与模型组比较,加味左归丸组与模型组比较,差异均有统计学意义(P<0.05),假手术组与加味左归丸组比较,差异无统计学意义(P>0.05)。结论:加味左归丸能够部分逆转OVX诱导的骨质疏松小鼠的骨丢失和骨微观结构的损伤,为补肾益气活血法治疗骨质疏松提供了实验依据。Objective:To explore the effect of Modified Zuo-gui-wan on the bone microstructure of ovariectomized osteoporosis model mice and its possible mechanism of action,in order to provide a basis for the treatment of osteoporosis with the method Journal of Sichuan of Traditional Chinese Medicine of Bu-shen-yi-qi-huo-xue.Method:218-week-old C57BL/6J female mice were randomly divided into a model group,a sham surgery group,and Modified Zuo-gui-wan group,with 7mice in each group.The model group and Modified Zuo-gui-wan group mice established osteoporosis models by removing both ovaries,while the sham surgery group mice removed an equal amount of adipose tissue.The Modified Zuo-gui-wan group was gavaged with Modified Zuo-gui-wan solution,while the model group and sham surgery group were gavaged with an equal amount of distilled water.After 8weeks of administration,bone tissue images were collected using mouse Micro CT combined with HE staining tissue sectioning technology to quantitatively analyze the trabecular bone and evaluate the effect of Modified Zuo-gui-wan on bone microstructure.Result:Observation of microstructure of mouse femur:The overall differences in bone volume fraction(BV/TV,%),trabecular thickness(Tb.Th,mm),and trabecular separation(Tb.Sp,mm)among the three groups of mice were statistically significant(P<0.05);The difference in bone volume fraction,trabecular thickness,and trabecular separation between the sham surgery group and the model group was statistically significant(P<0.05);Compared with the model group,the difference in bone volume fraction and trabecular thickness between the Modified Zuo-gui-wan group and the model group was statistically significant(P<0.05),while there was no statistically significant difference in trabecular separation(P>0.05);There was no statistically significant difference in bone volume fraction,trabecular thickness,and trabecular separation between the Modified Zuo-gui-wan group and the sham surgery group(P>0.05).Observation results of femoral tissue morphology in mice:Ov
