摘  要:小麦胚芽是小麦中最珍贵的部分,富含黄酮类化合物、谷胱甘肽以及小麦胚芽凝聚素等多种有益物质。在我们的生活中,小麦胚芽被利用得还不够充分。因此,研究采用乙醇回流法对麦胚中总黄酮进行了提取。采用紫外分光光度法,使用显色剂,建立芦丁标准曲线。通过开展单因素和正交试验优化,最终确定最佳条件:料液比为1∶40 g/mL,乙醇体积分数为70%,提取时间为2 h,提取温度为70℃,总黄酮的提取率为0.325 0%。Wheat germ is the most valuable part of wheat and is rich in many beneficial substances,such as flavonoids,glutathione and wheat germ coagulant.Nowadays,wheat germ is not more fully utilized except as animal feed in our life.Therefore,total flavonoids in wheat germ were extracted by the test using ethanol reflux method.A rutin standard curve was constructed by UV spectrophotometric method using chromogenic agent.The extraction criteria of total flavonoids in wheat germ were optimized by single-factor experiment and orthogonal experiment.Finally,the best extraction condition was obtained:the material-liquid ratio was 1∶40 g/mL,the volume fraction of ethanol was 70%,the extraction time was 2 h,and the extraction temperature was 70 ℃.The extraction rate of total flavonoids could reach 0.325 0% under this extraction condition.
