摘  要:通过实验分析亚甲基蓝与肝素的络合关系,讨论反应时间、溶液pH值、蛋白质的影响,建立一种快速、有效地检测肝素效价的方法。试验确定了分析波长为664 nm,且在0.09~0.54 IU/mL范围内的肝素与10 mg/L亚甲基蓝呈现良好的线性关系。反应时间在30~180 min范围内具有良好的稳定性;且该方法在pH=5~11范围内,1~30 mg/L牛血清蛋白(BSA)的干扰下仍保持较好的稳定性。对比实验中亚甲基蓝测肝素的方法与羊血浆法相差不大。该肝素检测方法简单、快速、经济、有效。By experimentally analyzing the complex relationship between methylene blue and heparin,discussing the effects of reaction time,solution pH value and protein,a rapid and effective method for detecting heparin titer was established.The experiment determined that heparin with an analytical wavelength of 664 nm and a range of 0.09~ 0.54 IU/mL showed a good linear relationship with 10 mg/L methylene blue.The reaction time had good stability in the range of 30~180 min.Moreover,the method maintained good stability under the interference of 1~30 mg/L bovine serum protein(BSA) in the range of pH was 5~11.The method of measuring heparin in methylene blue in the comparison experiment was not much different from the sheep plasma method.This method of heparin testing is simple,rapid,cost-effective and effective.
