摘  要:为了探究不同电荷对替米考星脂质体(TMS-Lip)靶向金黄色葡萄球菌的影响,试验制备带有不同电荷的TMS-Lip并对其进行表征,测定带有不同电荷TMS-Lip的抑菌圈直径、最小抑菌浓度,制备TMS-Lip卡波姆934凝胶,研究其对小鼠伤口感染模型的治疗效果。结果表明:研究制得具有不同电荷的三组(A、B、C组)TMS-Lip,三组TMS-Lip的Zeta电位分别为(-8.39±0.22)mV、(-20.30±0.67)mV、(-32.40±0.46)mV。随着Zeta电位的下降,脂质体的粒径从146.6 nm降低至91.1 nm,且在体外抗菌活性试验中的抑菌能力有所降低;但在体内动物试验中,随着Zeta电位的下降,A、B、C组小鼠的伤口愈合率先升高后降低,以带有适中负电荷的B组对小鼠伤口感染模型的治疗效果最佳。说明不同电荷对脂质体靶向金黄色葡萄球菌的影响存在体内外差异,在实际应用时,对脂质体Zeta电位的调整应多方位考虑与验证。In order to investigate the effects of different charges on tilmicosin liposomes(TMS-Lip)targeting Staphylococcus aureus,in the experiment,TMS-Lip with different charges were prepared and characterized;the diameter of inhibition zone and minimum inhibitory concentration with different charges of TMS-Lip were determined,and TMS-Lip carbomer 934 gel was prepared to study its therapeutic effect on a mouse wound infection model.The results showed that three groups(groups A,B,C)of TMS-Lip with different charges were produced with Zeta potentials of(-8.39±0.22)mV,(-20.30±0.67)mV,and(-32.40±0.46)mV,respectively.With the decrease of Zeta potentials,the particle sizes of liposomes decreased from 146.6 nm to 91.1 nm,and the inhibitory ability was decreased in the in vitro antimicrobial activity test.However,in the in vivo animal experiment,with the decrease of Zeta potentials,the wound healing of mice in groups A,B,C was firstly increased and then decreased,and group B with moderate negative charge had the best therapeutic effect on the wound infection model of mice.The results suggested that there was an in vivo and in vitro difference in the effect of different charges on liposomes targeting Staphylococcus aureus.In practical application,the adjustment of liposome Zeta potentials should be considered and verified in many aspects.
