摘 要:为了研究未性成熟(1岁)与性成熟(10岁)蒙古马睾丸组织形态及生精上皮细胞的区别,试验采集1岁和10岁蒙古马睾丸组织制作石蜡切片并进行H.E.染色,观察睾丸组织结构、生精上皮细胞形态及数量;采用实时荧光定量PCR方法和免疫组织化学染色法检测生精上皮细胞相关标志基因(FGFR3、GDNF、KITLG、ITGA6、DAZL)的差异表达情况。结果表明:1岁蒙古马睾丸组织中曲细精管未出现空腔结构,10岁则出现明显管腔结构;1岁蒙古马睾丸组织中曲细精管直径及面积分别为(59.51±4.35)μm和(2795.89±429.39)μm^(2),与10岁[(276.69±20.88)μm和(60459.26±8858.68)μm^(2)]相比差异极显著(P<0.01);1岁蒙古马睾丸曲细精管内部未见精子细胞,且生精上皮细胞数量为(27.67±5.09)个,与10岁[(239.90±21.07)个]相比差异极显著(P<0.01);10岁蒙古马睾丸曲细精管中央出现精子细胞。10岁蒙古马睾丸组织中精原细胞相关标志基因(ITGA6、DAZL、FGFR3)及支持细胞相关标志基因(GDNF、KITLG)的相对表达量极显著高于1岁蒙古马(P<0.01)。DAZL、GDNF基因在1岁和10岁蒙古马睾丸组织曲细精管中均呈阳性表达,且10岁蒙古马的阳性表达情况明显优于1岁。说明随着生殖系统的不断发育,精子发生的持续进行,未性成熟与性成熟蒙古马睾丸在组织形态及生精上皮细胞数量上存在极显著差异。To study the differences of testicular tissue morphology and spermatogenic epithelial cells between immature(1-year-old)and sexually mature(10-year-old)Mongolian horses,the experiment collected testicular tissues of 1 year old and 10 years old Mongolian horses,and made paraffin sections and performed H.E.The tissue structure,morphology and number of spermatogenic epithelial cells were observed in the testis;subsequently,real-time fluorescence quantitative PCR and immunohistochemical staining were used to detect the differential expression of seminiferous epithelial cell-related marker genes(FGFR3,GDNF,KITLG,ITGA6,DAZL).The results showed that there was no cavity structure in the curved fine ductus in the testicular tissue of the 1-year-old Mongolian horse.The diameter and area of the seminiferous tubules at(59.51±4.35)μm and(2795.89±429.39)μm^(2)were significantly different from those at 10-year-old(276.69±20.88)μm and(60459.26±8858.68)μm^(2)(P<0.01).No sperm cells were found in the tubule of the one-year-old Mongolian horse testis,and the number of spermatogenic epithelial cells(27.67±5.09)was significantly different from that of the 10-year-old(239.90±21.07)(P<0.01),and sperm cells appeared in the central tubule of the 10-year-old.The expression levels of spermatogonia-related marker genes(ITGA6,DAZL,FGFR3)and supporting cell-related marker genes(GDNF,KITLG)in testicular tissue of 10-year-old Mongolian horses were significantly higher than those of 1-year-old(P<0.01).DAZL and GDNF were expressed positively in the convoluted spermatotubule of the testicular tissue of Mongolian horses at 1 and 10 years old,and the positive expression of DAZL and GDNF at 10 years old was significantly higher than that at 1 year old.These results indicated that with the continuous development of the reproductive system and the continuous progress of spermatogenesis,there were significant differences in tissue morphology and the number of spermatogenic epithelial cells between the immature and sexually mature Mongolian hor
