摘 要:试验旨在探究梅花鹿、驯鹿与麋鹿机体基因表达模式的差异。以3岁龄、健康雄性梅花鹿、驯鹿与麋鹿各5头为研究对象,置于同一鹿场饲养,所有动物每日饲喂相同全混合饲粮,每天饲喂2次,试验预饲期7 d,正试期21 d。在正试期结束后的第1天晨饲后3 h采用盐酸赛拉嗪麻醉梅花鹿、驯鹿和麋鹿,经颈静脉采血并提取血液RNA。采用RNA-seq技术比较3种动物血液基因表达的差异。主成分分析(Principal components analysis,PCA)结果表明:梅花鹿、驯鹿与麋鹿血液的基因表达存在显著差异。与驯鹿相比,梅花鹿血液中共鉴定到2932个差异表达基因(Differently expressed genes,DEGs),被富集到超长链脂肪酸的代谢过程等GO功能及过氧化物酶体增殖物激活受体(Peroxisome proliferator-activated receptor,PPAR)信号通路等KEGG通路;与麋鹿相比,梅花鹿血液中共鉴定到6765个DEGs,被富集到脂蛋白分解代谢等GO功能及核糖体等KEGG通路;与麋鹿相比,驯鹿血液中共鉴定到5930个DEGs,被富集到蛋白质分泌等GO功能及不饱和脂肪酸的生物合成等KEGG通路。以上结果表明,在相同饲养条件下,梅花鹿、驯鹿与麋鹿机体的基因表达模式存在显著差异,结果可为理解鹿科动物的基因表达模式提供参考。This study was aimed to explore the differences in gene expression patterns among sika deer,reindeer and Père David's deer.In the experiment,five 3-year-old,healthy male sika deer,reindeer,and Père David's deer were kept on the same farm.All animals were fed with the same whole mixed diet every day.The pre-trial period lasted for 7 days and the full trial period for 21 days.On the 1st day after trial period finished,sika deer,reindeer and Père David's deer were anaesthetized using xylazine hydrochloride after morning feeding.RNA-seq technique was used to compare the differences in blood gene expression among them.The principal components analysis(PCA)results showed that the blood gene expression patterns were significantly different among sika deer,reindeer and Père David's deer.Compared with reindeer,2932 differently expressed genes(DEGs)were identified in the blood of sika deer,which were enriched in GO terms such as very long-chain fatty acids metabolic process and KEGG pathways such as peroxisome proliferatoractivated receptor(PPAR)signaling pathway.Compared with Père David's deer,6765 DEGs were identified in the blood of sika deer,which were enriched in GO terms such as lipoprotein metabolic process and KEGG pathways such as ribosome.Compared with Père David's deer,5930 DEGs were identified in the blood of reindeer,which were enriched in GO terms such as protein secretion and KEGG pathways such as biosynthesis of unsaturated fatty acids.In conclusion,the body gene expression patterns of sika deer,reindeer and Père David's deer were significantly different under the same feeding condition,which may provide a reference for comprehending the gene expression patterns of cervidea.
