摘 要:目的探讨白藜芦醇(Resveratrol,RSV)通过双调蛋白(Amphiregulin,AREG)/表皮生长因子受体(Epider-mal Growth Factor Receptor,EGFR)通路减轻输尿管梗阻后继发肾损伤的保护机制。方法选取2023年2—9月牡丹江医学院实验动物中心饲养30只SD大鼠为研究对象,依据随机数字表法分为对照组、单侧输尿管梗阻(Unilateral Ureteral Obstruction,UUO)组和UUO+白藜芦醇组,每组10只。对比3组小鼠的氧化应激指标[丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Superoxide Dismutase,SOD)]、AREG及EGFR的mRNA表达水平,分析AREG/EGFR通路标志物和肾纤维化(Renal Interstitial Fibrosis,RIF)相关指标[纤连蛋白(Fibronectin,FNO)、α-平滑肌肌动蛋白(α-smooth Muscle Actin,α-SMA)、E-黏连蛋白(E-cadherin)]。结果与对照组比较,UUO组的α-SMA和FNO蛋白表达水平升高,E-cadherin蛋白表达降低,MDA表达水平增高,SOD表达水平降低,AREG、EGFR蛋白表达增高,差异有统计学意义(P均<0.05)。与UUO组比较,UUO+RSV组的α-SMA和FNO蛋白表达下降,E-cadherin蛋白表达升高,MDA表达水平降低,SOD表达水平增高,AREG、EGFR蛋白表达下降,差异有统计学意义(P均<0.05)。结论白藜芦醇通过调控AREG/EGFR通路改善UUO大鼠RIF及氧化应激。Objective To explore the protective mechanism of resveratrol(RSV)in alleviating secondary renal injury after ureteral obstruction through the amphiregulin(AREG)/epidermal growth factor receptor(EGFR)pathway.Meth-ods Thirty SD rats raised in the Experimental Animal Center of Mudanjiang Medical College from February to Septem-ber 2023 were selected as the research objects and divided into control group according to the random number table method,unilateral ureteral obstruction(UUO)group and UUO+resveratrol group,with 10 rats in each group.The mRNA expression levels of malondialdehyde(MDA),superoxide dismutase(SOD),AREG and EGFR in the three groups were compared.The markers of AREG/EGFR pathway and related indicators of renal interstitial fibrosis(RIF)[fibronectin(FNO),α-smooth muscle actin,renal interstitial fibrosis(RIF)]were analyzed.α-SMA),E-cadherin].Re-sults Compared with the control group,the protein expression levels ofα-SMA and FNO in UUO group were in-creased,the protein expression of E-cadherin was decreased,the MDA expression level was increased,the SOD ex-pression level was decreased,and the protein expressions of AREG and EGFR were increased,the differences were statistically significant(all P<0.05).Compared with UUO group,α-SMA and FNO protein expression decreased,E-cadherin protein expression increased,MDA expression level decreased,SOD expression level increased,and AREG and EGFR protein expression decreased in UUO+RSV group,with statistical significance(all P<0.05).Conclusion Resveratrol can improve RIF and oxidative stress in UUO rats by regulating AREG/EGFR pathway.
