摘 要:目的筛选问荆止血作用的活性部位,初步探讨其止血作用机制。方法制备问荆石油醚、乙酸乙酯、正丁醇、水部位提取物,将昆明小鼠随机分成14组:空白组、云南白药组,水部位组、正丁醇部位组、乙酸乙酯部位组、石油醚部位组(各极性部位剂量:6、4、2 g/kg),连续给药7 d后,采用断尾法和眼眶取血法测定出血时间(BT)、凝血时间(CT);采用自动凝血仪测定凝血四项:凝血酶原时间(PT)、凝血酶时间(TT)、活化部分凝血酶时间(APTT)、纤维蛋白原(FIB)浓度。结果与空白组比较,乙酸乙酯部位(剂量:6、4、2 g/kg)组、石油醚部位(剂量:6 g/kg)组可显著缩短小鼠的BT、CT、PT、APTT(P<0.01);石油醚部位(剂量:4 g/kg)组可缩短小鼠的BT、CT、PT、APTT(P<0.05),正丁醇部位(剂量:6、4、2 g/kg)组、水部位(剂量:6、4、2 g/kg)组及石油醚部位(剂量:2 g/kg)组对小鼠的BT、CT、PT、APTT均无明显影响(P>0.05),各提取部位对小鼠的TT及FIB浓度均无明显影响(P>0.05)。结论问荆止血作用活性部位为石油醚部位、乙酸乙酯部位,作用机制与激活内、外源性凝血系统有关。Objective To screen the active fractions of Equisetum arvense L.for hemostasis,so as to investigate its hemostatic mechanism.Methods The extracts of petroleum ether,ethyl acetate,n-butanol and water fractions of Equisetum arvense L.were prepared,and Kunming mice were randomly divided into 14 groups:the blank group,the Yunnan Baiyao Powder group,the water fraction group,the n-butanol fraction group,the ethyl acetate fraction group and the petroleum ether fraction group(dosage of each polar fraction:6,4 and 2 g/kg).After 7 days of continuous administration,the bleeding time(BT)and coagulation time(CT)were measured by tail-cutting method and orbital blood collection method.Four coagulation parameters were measured using an automatic coagulation analyzer,which were prothrombin time(PT),thrombin time(TT),activated partial thromboplastin time(APTT)and fibrinogen(FIB)concentration.Results Compared with the blank group,the ethyl acetate fraction(dosages:6,4,2 g/kg)group and the petroleum ether fraction(dosage:6 g/kg)group could remarkably shorten the BT,CT,PT and APTT of mice(P<0.01).The petroleum ether fraction(dosage:4 g/kg)group could shorten the BT,CT,PT and APTT of mice(P<0.05).The n-butanol fraction(dosages:6,4,2 g/kg)group,the water fraction(dosages:6,4,2 g/kg)group,and the petroleum ether fraction(dosage:2 g/kg)group had no remarkable impacts on the BT,CT,PT,and APTT of mice(P>0.05).Each extraction fraction had no remarkable impact on the TT and FIB concentrations in mice(P>0.05).Conclusion The active fractions of Equisetum arvense L.for hemostasis are petroleum ether fraction and ethyl acetate fraction,and the function mechanism is related to the activation of endogenous and exogenous coagulation systems.
